乳腺癌组织中CBX2表达水平变化及其对PI3K/Akt信号通路的作用机制 被引量:4

摘要 目的:探讨乳腺癌患者色素框同源物2(CBX2)表达水平变化及其对PI3K/Akt信号通路的作用机制。方法:选取2015年1月—2016年1月延安市人民医院收治的155例乳腺癌患者作为研究对象,比较乳腺癌组织和癌旁正常组织中CBX2的表达水平,分析乳腺癌组织中CBX2表达与临床病理参数间的关系,比较不同CBX2表达水平的乳腺癌患者生存期,并采用Cox比例风险回归模型对乳腺癌患者生存期影响因素进行分析。构建CBX2表达载体转染乳腺癌细胞系,分别对转染组(转染含CBX2基因的载体)、阴性对照组(转染空质粒)和未转染组(未转染任何质粒)细胞ALK、PI3K、p-ALK和p-PI3K蛋白的表达量进行检测,分析CBX2对PI3K/Akt信号通路相关蛋白表达的影响。结果:CBX2蛋白在乳腺癌组织中的高表达率显著高于癌旁正常组织(P<0.05);乳腺癌组织中CBX2表达与肿瘤直径、TNM分期及淋巴结转移相关(P<0.05);CBX2高表达组乳腺癌患者5年生存率(75.3%)显著低于CBX2低表达组乳腺癌患者(90.3%),差异有统计学意义(P<0.05);Cox单因素和多因素分析结果显示肿瘤大小、TNM分期、淋巴结转移及CBX2表达水平是乳腺癌预后的影响因素。Western blot结果显示CBX2转染组p-ALK和p-PI3K蛋白表达水平显著高于未转染组和阴性对照组(P<0.05)。结论:CBX2可能通过PI3K/Akt信号通路相关蛋白参与乳腺癌的发生和发展,并影响乳腺癌的预后。 Objective:To investigate the changes of CBX2 expression and its mechanism on PI3K/Akt signaling pathway in patients with breast cancer.Methods:During January 2015 to January 2016,155 patients with breast cancer in Yan'an People's Hospital were selected as the subjects,and the expression levels of CBX2 in breast cancer tissues and adjacent normal tissues were compared.The relationship between CBX2 expression and clinicopathological parameters in breast cancer was analyzed,and the survival time of breast cancer patients with different CBX2 expression levels was compared,and the influencing factors of survival time of breast cancer patients were analyzed by COX proportional risk regression model.To construct CBX2 expression vector transfected with breast cancer cell lines,and to detect the expression of ALK、PI3K、p-ALK and p-PI3K proteins in transfection group(transfected with containing CBX2 gene vector),negative control group(transfected with empty plasmid)and untransfected group(not transfected with any plasmid),To analyze the effect of CBX2 on PI3K/Akt signaling path-related protein expression.Results:The high expression rate of CBX2 protein in breast cancer tissue was significantly higher than that in normal tissues adjacent to cancer(P<0.05).The expression of CBX2 in breast cancer was significantly correlated with tumor diameter,TNM stage and lymph node metastasis(P<0.05).The 5-year survival rate(75.3%)of patients with breast cancer in CBX2 high-expression group was significantly lower than that in CBX2 low-expression group(90.3%).Cox single and multivariate analysis showed tumor size,TNM stage,lymph node metastasis and CBX2.The expression level of p-ALK and p-PI3K protein in CBX2 transfection group was significantly higher than that in control group and negative control group(P<0.05).Conclusions:CBX2 may participate in the development and development of breast cancer through PI3K/Akt signaling pathway related proteins and affect the prognosis of breast cancer.
