动物双歧杆菌和罗伊氏乳杆菌改善肥胖小鼠糖脂代谢异常的研究 被引量:4

EFFECTS OF BIFIDOBACTERIUM CP-9 AND LACTOBACILLUS REUTERI ON GLUCOSE AND LIPID METABOLISM IN OBESE MICE

摘要 目的了解动物双歧杆菌(CP-9)和罗伊氏乳杆菌(Lactobacillus reuteri)干预对肥胖小鼠糖脂代谢异常的改善作用。方法选择6w龄雄性C57BL/6J小鼠高脂饲料喂养8w,挑选造模成功的肥胖小鼠24只分为3组持续高脂饲料喂养,动物双歧杆菌组(CP-9,109cfu/d)和罗伊氏乳杆菌组(L.reuteri,109cfu/d)给予对应益生菌干预10w。另随机选择同日龄正常小鼠8只为空白对照。干预期结束后,结合体重、血糖、血脂、肝脏FAS基因和PPARγ基因蛋白表达等指标来评估益生菌干预对肥胖小鼠糖脂代谢的影响。结果CP-9和L.reuteri干预结束时体重分别为(37.26±1.48)、(35.78±1.20)g,明显低于造模对照组(46.33±2.84)g。CP-9和L.reuteri干预9w末空腹血糖分别为(5.28±0.96)、(5.52±0.77)mmol/L,明显低于造模对照组(7.19±1.23)mmol/L;干预结束时,糖化血红蛋白百分比中位数为[3.60(3.25,4.35)]和[3.50(3.40,3.65)],明显低于造模对照组[4.50(4.20,4.90)];口服糖耐量实验曲线下面积(area under the curve,AUC)分别为1858.89±274.21和1733.36±138.58,均低于造模对照组1899.00±274.80;血清胰岛素(27.96±3.43)、(23.70±1.78)mIU/L,明显低于造模对照组(32.96±2.02)mIU/L;胰岛素抵抗指数中位数为[6.65(4.92,7.63)]和[5.70(5.06,6.07)],明显低于造模对照组[10.43(9.38,13.47)];胰岛β细胞功能指数中位数为[223.00(189.00,734.67)]和[251.98(224.91,279.46)],明显高于造模对照组[167.99(130.48,177.48)];血清低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)水平为(0.70±0.22)、(0.81±0.13)mmol/L,明显低于造模对照组(0.94±0.24)mmol/L;血清总甘油三酯(triglyceride,TG)水平为(1.07±0.21)、(1.21±0.10)mmol/L,明显低于造模对照组(1.73±0.37)mmol/L。CP-9和L.reuteri上调肝脏FAS基因蛋白表达,下调肝脏PPARγ基因蛋白表达。结论CP-9和L.reuteri干预可有效改善肥胖小鼠糖脂代谢异常状态,通过调节肝脏FAS和PPARγ基因的蛋白表达,降低血糖异常升高,改善胰岛 Objective To investigate the effects of bifidobacterium(CP-9)and lactobacillus royale(Lactobacillus reuteri)on glucose and lipid metabolism in obese mice.Methods Six-week-old male C57 BL/6 J mice were fed a high-fat diet for 8 weeks to establish an obesity model.Mice that had been properly modeled were selected and divided into three groups(n=8):the model control group(high-fat diet),blank control group(normal diet),and CP-9(high-fat diet+bifidobacterium CP-9109 cfu/d)and L.reuteri(high-fat diet+Lactobacillus reuteri 109 cfu/d)groups.All the three groups were subjected to 10 weeks of probiotics intervention.In addition,another 8 normal mice of the same age were randomly selected as the blank control group(ND).At the end of 10 weeks,the effects of probiotic intervention on blood glucose and lipids in obese mice were evaluated based on fasting blood glucose,glycosylated hemoglobin,oral glucose tolerance,insulin and blood lipids.Results At the end of intervention with CP-9 and L.reuteri,body weight was(37.26±1.48)g and(35.78±1.20)g respectively,significantly lower than that of the model control group.The fasting blood glucose was(5.28±0.96)mmol/L and(5.52±0.77)mmol/L respectively,significantly lower than that of the model control group.HbA1c%was[3.60(3.25)4.35]and[3.50(3.40)3.65]respectively,which was significantly lower than that of the model control group.The area under the curve of oral glucose tolerance test was(1858.89±274.21)and(1733.36±138.58)respectively,which was lower than that of the model control group.The insulin resistance index was[6.65(4.92,7.63)]and[5.70(5.06,6.07)],which was significantly lower than that of the model control group,so were the isletβcell function indexes[223.00(189.00734.667)]and[251.98(224.91279.46)].The levels of serum low density lipoprotein cholesterol(LDL-C)(0.70±0.22)and(0.81±0.13)mmol/L,and serum total triglyceride(TG)(1.07±0.21)and(1.21±0.10)mmol/L were also significantly decreased.CP-9 and L.reuteri up-regulated the expression of FAS protein and down-regulated th
