摘要
小麦胚芽含有丰富的营养物质,利用小麦胚芽蛋白制备生物活性肽具有重要意义。以DPPH自由基清除率为检测指标,通过酶解小麦胚芽蛋白产生活性肽。通过筛选发现碱性蛋白酶酶解小麦胚芽蛋白效果最佳,通过单因素法和正交试验法确定的最佳酶解条件为底物浓度8%、酶解温度55℃、pH 7.5、加酶量3000 U/g及酶解时间2 h。在此条件下酶解液的DPPH自由基清除率达88.91%±1.5%,并发现此时酶解液的粗品中含有1.28 mg/mL的小麦胚芽肽。通过葡聚糖G-50凝胶层析分离得到3种组分,它们对DPPH自由基清除能力分别为53.24%±1.9%,77.74%±3.7%和50.77%±2.4%。利用葡聚糖G-25凝胶层析对活性最高的组分进一步纯化,最终得到单一蛋白组分,其总抗氧化性能力为0.511 mmol/L Trolox当量,清除羟基自由基的能力为56.45 U/mL,抗超氧阴离子能力为45.31 U/L。
Wheat germ is rich in nutrients,and it is very important to use wheat germ protein to prepare bioactive peptides.Aim to produce bioactive peptides by enzymatic hydrolysis of wheat germ protein,with the DPPH free radical scavenging rate used as a detection index.It was found that alkaline protease had the best effect on enzymatic hydrolysis of wheat germ protein.The condition for enzymatic hydrolysis by single factor experiment and orthogonal experiment was optimized.The optimized hydrolysis condition was that substrate concentration of 8%,temperature of enzymatic hydrolysis of 55℃,pH 7.5,adding enzyme amount of 3000 U/g and enzymatic hydrolysis time of 2 h.Under this condition,the DPPH radical-scavenging rate of the enzymolysis solution was as high as 88.91%±1.5%,and the crude product of the enzymolysis solution contained 1.28 mg/m L wheat germ peptide.The hydrolysate was separated and purified by dextran G-50 gel chromatography to obtain three components,and their DPPH radical-scavenging rates were 53.24%±1.9%,77.74%±3.7%and 50.77%±2.4%,respectively.The highest component was further purified by dextran G-25 gel chromatography,and a single protein component was obtained.Its total antioxidant capacity was a trolox equivalent antioxidant capacity of 0.511 mmol/L Trolox;Its hydroxyl radical scavenging capacity was 56.45 U/mL;Its superoxide anion resistance capacity was 45.31 U/L.
