树突状细胞介导心肌梗死大鼠特异性细胞免疫对心室功能改善的作用

Improving effect of specific cellular immunity mediated by dendritic cells on ventricular function in rats with acute myocardial infarction

摘要 目的探讨树突状细胞(DC)介导心肌梗死(AMI)大鼠特异性细胞免疫对心室功能改善的作用。方法购自北京医疗器械检验所(动物实验中心)清洁级雄性大鼠24只,构建AMI手术模型后将其随机分成A、B、C三组,每组各8只,A组为假手术大鼠、B组为AMI大鼠、C组为接受DC干预的AMI大鼠。记录大鼠生存情况;每组4只用于DC、血流动力学指标和炎性因子检测,另4只用于HE染色检测细胞炎性浸润、细胞凋亡及心肌梗死情况。结果C组生存率高于A组(P<0.05);AMI后第3 d可检出DC相关标志物,第7 d达高峰,第14 d后下降;C组CD40、CD80、CD86、MHCⅠ、MHCⅡ表达水平高于A、B组(P<0.05);流式分析结果显示,DC可获取CD4^(^(+))CD25^(+)纯度>90%,且可有效诱导Treg细胞群;Treg在大鼠心脏和淋巴结中均有表达,且C组表达水平高于A、B组(P<0.05);C组动力学指标、炎性因子水平优于A、B组(P<0.05);C组心肌梗塞面积、胶原体积分数(CVF)低于A、B组(P<0.05),且基质金属蛋白酶2(MMP-2)活性高于A、B组(P<0.05);C组炎性细胞计数、细胞凋亡<A、B组(P<0.05)。结论DC可激活AMI后特异性细胞免疫,修复炎性细胞,进一步改善心室重塑及功能,提高AMI大鼠生存率。 Objective To investigate the improving effect of specific cellular immunity mediated by dendritic cells(DC)on ventricular function in rats with acute myocardial infarction(AMI).Methods Male rats(n=24)were randomly divided into group A(sham-operation group),group B(AMI group)and group C(AMI^(+)DC group,each n=8)after AMI model established.The rat survival situation was recorded.The changes of DC,hemodynamic indexes and inflammatory factors were detected in 4 rats in each group,and inflammatory infiltration,apoptosis and myocardial infarction were detected by using HE staining in other 4 rats in each group.Results The survival rate was higher in group C than that in group A(P<0.05).DC-related biomarkers were detected on the 3rd d after AMI,reached peak on the 7th d and decreased on the 14th d.The expressions of CD40,CD80,CD86,MHC Ⅰ and MHCⅡwere higher in group C than those in group B and group A(P<0.05).The results of flow cytometry analysis showed that DC could obtain CD4^(+)CD25^(+)with purity greater than 90%,and induce effectively Treg cell population.The expression of Treg could be observed in rat heart and lymph node,which was higher in group C than that in group B and group A(P<0.05).The hemodynamic indexes and inflammatory factors were superior in group C to those in group B and group A(P<0.05).The area of myocardial infarction and collagen volume fraction(CVF)were lower(P<0.05),and activity of matrix metalloproteinase 2(MMP-2)was higher(P<0.05)in group C than those in group B and group A.The count of inflammatory cells and apoptosis rate were lower in group C than those in group B and group A(P<0.05).Conclusion DC can activate specific cellular immunity,repair inflammatory cells,further improve ventricular remodeling and function,and improve survival rate of AMI rats.
