中药软肝饮对HBV转基因小鼠iNKT细胞及其表面PD-1、PD-L1表达的影响 被引量:1

摘要 目的:研究中药软肝饮对HBV转基因小鼠iNKT细胞及其表面PD-1、PD-L1表达的影响,探讨中药软肝饮对HBV转基因小鼠抗病毒的作用机制。方法:将50只雌雄各半HBV转基因小鼠随机分为治疗组(高、中、低剂量软肝饮)、模型组、阳性对照组(恩替卡韦分散片)等5组,另用10只非HBV转基因小鼠作为空白对照。药物干预灌胃4周后处死小鼠,观察各组小鼠外周血及肝脏中iNKT细胞及其表面PD-1、PD-L1的表达。结果:与空白对照组相比,模型组小鼠外周血iNKT细胞表达明显降低,iNKT细胞表面PD-1、PD-L1的表达水平明显增高,差异有显著性意义(P<0.01);与模型组相比,治疗后软肝饮高剂量组和阳性对照组小鼠外周血iNKT细胞表达明显增高,iNKT细胞表面PD-1、PD-L1的表达水平有一定程度的降低,差异有统计学意义(P<0.01或P<0.05);软肝饮中剂量组小鼠外周血iNKT细胞表达有一定增加,有一定差异性(P<0.05),而iNKT细胞表面PD-1、PD-L1的表达水平无明显变化,无明显差异性;软肝饮低剂量组小鼠外周血iNKT细胞表达及iNKT细胞表面PD-1、PD-L1的表达无明显变化,无明显差异性(P>0.05)。HBV转基因小鼠肝脏中细胞iNKT的表达明显低于空白对照组,iNKT细胞表面PD-1、PD-L1的表达明显高于空白对照组,均有统计学意义(P<0.05);结论:中药软肝饮调控HBV转基因小鼠的免疫机制可能通过阻断iNKT细胞上PD-1/PD-L1信号通路,调控iNKT细胞功能发挥抗HBV作用。 Objective:To study the effect of Ruanganyin on the expression of PD-1 and PD-L1 on iNKT cells and their surface in HBV transgenic mice,and to explore the antiviral mechanism of Ruanganyin on HBV transgenic mice.Methods:A total of 50 HBV transgenic mice(half male and half female)were randomly divided into treatment group(high,medium and low dose Ruanganyin),model group,positive control group(entecavir dispersible tablets)and ten non HBV transgenic mice as blank control.The mice were killed after four weeks of drug intervention.The expression of iNKT cells and PD-1,PD-L1 in peripheral blood and liver of each group were observed.Results:compared with the blank control group,the expression of iNKT cells in the model group was significantly decreased,and the expression levels of PD-1 and PD-L1 on the surface of iNKT cells were significantly increased(P<0.01);compared with the model group,the expression of iNKT cells in the high-dose Ruanganyin group and the positive control group was significantly increased after treatment The expression levels of PD-1 and PD-L1 on the surface of iNKT cells decreased to a certain extent(P<0.01/P<0.05);the expression of iNKT cells in the middle dose group of Ruanganyin increased to a certain extent,with a certain difference(P<0.05),while the expression levels of PD-1 and PD-L1 on the surface of iNKT cells had no significant change,and there was no significant difference;in the low-dose group of Ruanganyin,the expression of iNKT cells and PD-1,PD-L1 on the surface of iNKT cells were not significantly different and no significant difference(P>0.05);the expression of iNKT surface in HBV transgenic mice was significantly lower than that in the blank control group,and the expression of PD-1 and PD-L1 on the surface of iNKT cells was significantly higher than that in the blank control group(P<0.05).After four weeks of treatment,compared with the model group,the expression of iNKT cells in the liver cells of HBV transgenic mice in the high,medium,and low dose groups of Ruanganyin and the posi
