摘要
目的:观察复方黄柏液对溃疡性结肠炎(ulcerative colitis, UC)大鼠相关炎性因子及核转录因子-κB(nuclear factor-kappa B,NF-κB)和环氧合酶-2(cyclooxygenase-2,COX-2)蛋白表达的影响,探讨复方黄柏液对大鼠结肠黏膜的保护机制。方法:实验动物随机分为空白对照组、模型组、复方黄柏液组,采用免疫复合法制备UC大鼠模型,治疗14 d后,采用ELISA法检测白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumour necrosis factor-α,TNF-α)变化,Western Blot法检测NF-κB和COX-2蛋白表达。结果:与空白组对比,模型组IL-6和TNF-α含量升高(P<0.01),NF-κB和COX-2蛋白表达升高(P<0.01);与模型组对比,复方黄柏液组中IL-6和TNF-α含量降低(P<0.05),NF-κB和COX-2蛋白表达降低(P<0.05)。结论:复方黄柏液对UC大鼠的保护可能通过抗炎作用。
Objective: To observe the effects of Compound Cortex Phellodendri Fluid on inflammatory factors, nuclear factor-kappa B(NF-κB) and cyclooxygenase-2(cyclooxygenase-2, COX-2) protein expression in rats with ulcerative colitis(ulcerative colitis, UC), and explore the protective mechanism. Methods: The experimental animals were randomly divided into blank control group, model group and Compound Cortex Phellodendri Fluid group. The UC rat model was established by immune complex method. After 14 days of treatment, the contents of interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) were determined by ELISA method. Nuclear factor-κB and cyclooxygenase-2 were determined by Western Blot method. Results: Compared with the blank group, the contents of IL-6 and TNF-α and the expression of NF-κB and COX-2 protein in the model group increased, while the contents of IL-6 and TNF-α, and the expression of NF-κB and COX-2 protein in the Compound Cortex Phellodendri Fluid group decreased. Conclusion: The protective effect of Compound Cortex Phellodendri Fluid on UC rats may be produced through anti-inflammatory effect.
