摘要
目的:探讨急性髓细胞白血病(AML)模型小鼠B细胞易位基因1(BTG1)和Ikaros家族锌指转录因子1(IKZF1)表达水平,并进行相关性分析。方法:36只小鼠随机等分为模型A组、模型B组和对照组。模型A组和模型B组分别经尾静脉注射HL60细胞1×10^(7)/只、5×10^(6)/只,对照组注射等量0.9%氯化钠溶液。采用Western blot检测骨髓组织BTG1和IKZF1蛋白相对表达水平,并与体重、红细胞、血小板及白细胞CD33阳性率进行相关性分析。结果:模型A组和模型B组小鼠在接种第7、14、28天的体重、血小板低于对照组,红细胞高于对照组(均P<0.05),但模型A组与模型B组上述指标比较无统计学差异(均P>0.05)。模型A组和模型B组接种第28天白细胞CD33阳性率高于对照组,BTG1和IKZF1蛋白相对表达水平低于对照组(均P<0.05),但模型A组与模型B组上述指标比较无统计学差异(均P>0.05)。Pearson相关性分析显示,AML小鼠BTG1、IKZF1蛋白相对表达水平与白血病CD33阳性率、体重、红细胞、血小板均存在相关性(均P<0.05)。结论:BTG1、IKZF1蛋白在AML模型小鼠呈高表达,且表达水平与小鼠体重、外周血及肿瘤浸润程度存在相关性。
Objective:To investigate the expression of B cell translocation gene 1(BTG1)and Ikaros family zinc finger 1(IKZF1)in mouse model of acute myeloid leukemia(AML),and to analyze the correlation.Methods:36 mice were randomly divided into model A group,model B group and control group equally.model A group and model B group were injected with HL60 cells 1×10^(7)/mouse and 5×10^(6)/mouse respectively via tail vein,and the control group was injected with the same amount of normal saline.Western blot was used to detect the relative expression levels of BTG1 and IKZF1 proteins in bone marrow tissue,and the correlation analysis between the CD33 positive rate and body weight,red blood cells,platelets was carried out.Results:The body weight,platelets and red blood cells of the mice in the model A and model B groups on the 7th,14th,and 28th day of inoculation were lower than those in the control group(all P<0.05),but there was no statistical difference in the above indicators between the model A group and model B group(all P>0.05).The positive rate of CD33 on the 28th day of inoculation in the model A and model B groups was higher than that in the control group,and the relative expression levels of BTG1 and IKZF1 proteins were lower than those in the control group(all P<0.05),but there was no statistical difference in the above indicators between the model A group and the model B group(all P>0.05).Pearson correlation analysis showed that the relative expression levels of BTG1 and IKZF1 proteins in AML mice were correlated with the positive rate of CD33,body weight,red blood cells and platelets(all P<0.05).Conclusion:BTG1 and IKZF1 proteins are highly expressed in AML model mice,and their expression levels are correlated with mouse body weight,peripheral blood and tumor invasion.
