补肺健脾益肾方对COPD模型大鼠TLR4/MyD88/NF-κB通路相关因子的影响 被引量:9

摘要 目的:研究补肺健脾益肾方对大鼠慢性阻塞性肺疾病(COPD)模型Toll样受体4(TLR4)/髓样分化因子88(MyD88)/核转录因子κB(NF-κB)通路的影响。方法:采用随机数字表法将40只大鼠随机分为模型组、常规组、实验组、正常组,每组10只。取其干预12周后肺组织和支气管肺泡灌洗液,比较各组大鼠干预期间一般情况,比较各组大鼠TLR4 mRNA、MyD88 mRNA、NF-κB mRNA水平及肺组织形态学评分。结果:正常组大鼠在干预期间皮毛、活动度、饮水进食、体重增长趋势均优于其余三组,常规组及实验组上述表现均优于模型组。与正常组比较,模型组大鼠TLR4 mRNA、MyD88 mRNA、NF-κB mRNA水平均升高;与模型组比较,常规组、实验组以上指标均低,且实验组低于常规组,差异有统计学意义(均P<0.05)。与正常组比较,模型组大鼠肺组织上皮细胞变性坏死糜烂脱落、气道管壁黏膜充血水肿、气道管壁中性粒细胞浸润评分均升高,与模型组比较,常规组、实验组以上指标均降低,且实验组低于常规组,差异有统计学意义(均P<0.05)。结论:补肺健脾益肾方可通过抑制大鼠TLR4/MyD88/NF-κB通路相关因子表达改善COPD的气道炎症反应,提高治疗效果。 Objective:To study effect of Bufei Jianpi Yishen decoction on inhibition of Toll like receptor 4(TLR4)/myeloid differentiation factor 88(MyD88)/nuclear transcription factor-κB(NF-κB)pathway in rats with chronic obstructive pulmonary disease(COPD).Methods:40 rats were randomly divided into model group,routine group,experimental group and normal group with 10 rats in each group.The lung tissue and bronchoalveolar lavage fluid were taken after 12 weeks of intervention.The general conditions of rats in each group during intervention were compared.Levels of TLR4 mRNA,MyD88 mRNA,NF-κB mRNA and lung histomorphological score of rats in each group were compared.Results:During intervention period,fur,activity,drinking and eating,weight growth trend of the normal group were better than those of other three groups.The above performance of the routine group and the experimental group were better than that of the model group.Compared with the normal group,the TLR4 mRNA,MyD88 mRNA,NF-κB mRNA level in the model group increased significantly.The above indicators in the experimental group and the conventional group were lower than the model group,difference statistically significant(all P<0.05).Compared with the normal group,erosion of pulmonary epithelial cell necrosis,congestion and oedema of airway tube wall,and neutrophil infiltration scores of rats in the model group were significantly increased.The erosion of pulmonary epithelial cell necrosis,congestion and oedema of the airway tube wall,and neutrophil infiltration scores in the experimental group and the conventional group were lower than the model group,difference statistically significant(all P<0.05).Conclusion:Bufei Jianpi Yishen decoction can reduce airway inflammation induced by COPD and improve therapeutic effect by inhibiting expression of TLR4/MyD 88/NF-κB pathway related factors.
