复合丝素导管修复大鼠坐骨神经损伤的研究 被引量:1

摘要 目的:研究复合型丝素神经导管对大鼠坐骨神经损伤的修复效果,为复合丝素支架在周围神经损伤中的临床应用提供理论依据。方法:制备两种分子质量的丝素蛋白,并通过静电纺丝、编织、涂层等方法制备复合型丝素蛋白导管,将导管在兔子的背部皮下植入,设定相应时间点进行观察和统计分析。在大鼠坐骨神经缺损模型中,通过苏木精-伊红染色、组织免疫荧光染色和透射电镜等手段观察导管的生物安全性以及促坐骨神经再生能力。结果:在兔子背部皮下植入模型中,24周时导管表现出良好的降解趋势;在大鼠坐骨神经缺损模型中,导管无明显不良反应,表明该导管具有良好的生物安全性;12周时,导管能够促进轴突和髓鞘的形成从而促进坐骨神经再生,还能改善肌肉萎缩。结论:低分子丝素复合导管具有良好的生物安全性和降解性,并能促进大鼠坐骨神经的再生,为丝素在周围神经损伤修复领域的临床试验提供了理论依据。 Objective:The repair effect of combined silk fibroin nerve conduits on sciatic nerve injury in rats was studied,providing theoretical basis for the clinical application of combined silk fibroin scaffold in peripheral nerve injury.Methods:Two kinds of silk fibroin with different molecular weights were prepared,and then composite silk fibroin scaffolds were prepared by electrospinning,knitting and coating.The scaffolds were implanted subcutaneously on the back of rabbit,and the predetermined time point was set for observation and statistical analysis.In the rat model of sciatic nerve defect,the biological safety and the ability of promoting sciatic nerve regeneration were observed by means of hematoxylin-eosin staining,tissue immunofluorescence staining and transmission electron microscopy.Results:In rabbit dorsal subcutaneous implant model,the catheter showed a good tendency of degradation at 24 weeks.In the rat model of sciatic nerve defect,there was no obvious adverse reaction of the catheter,indicating that the catheter had good biosafety.At 12 weeks,catheterization promotes axon and myelin formation leading to sciatic nerve regeneration,and also improves muscle atrophy.Conclusion:The low molecular weight silk fibroin composite catheter has good biological safety and degradation,and can promote the regeneration of sciatic nerve in rats,which provides a theoretical basis for the clinical trials of silk fibroin in the field of peripheral nerve injury and repair.
