摘要
目的:探讨盐酸小檗碱对人口腔鳞癌细胞CAL-27、SCC-15增殖、迁移及侵袭能力的影响及可能机制。方法:通过CCK-8法检测不同浓度盐酸小檗碱对口腔鳞癌细胞增殖的影响并计算出合适的半数抑制浓度IC50进行后续实验;划痕、Transwell实验分别检测盐酸小檗碱对肿瘤细胞迁移及侵袭能力的影响。Western Blot检测MMP-2、MMP-9蛋白的表达情况;qRT-PCR检测各组细胞中侵袭迁移相关基因MMP-2和MMP-9 mRNA的表达情况。结果:盐酸小檗碱可显著抑制CAL-27细胞、SCC-15细胞的增殖,且呈浓度及时间依赖性;盐酸小檗碱可明显抑制口腔鳞癌细胞的迁移和侵袭能力;MMP-2、MMP-9 mRNA和蛋白表达均明显下降(P<0.05)。结论:盐酸小檗碱可有效抑制人口腔鳞癌细胞的增殖、迁移和侵袭能力,其可能是通过调控MMP-2、MMP-9的表达而发挥抗肿瘤作用。
Objective:To explore the effect of berberine hydrochloride on the proliferation, migration and invasion ability of human oral squamous cell carcinoma cells CAL-27 and SCC-15,and the potential mechanism.Methods:The effect of different concentrations of berberine hydrochloride on the proliferation of oral squamous cell carcinoma cells was detected by CCK-8 method, and the appropriate half inhibitory concentration IC50 was calculated for subsequent experiments.The effects of berberine hydrochloride on migration and invasion of the tumor cells were detected by scratch and Transwell assay.Western Blot was used to detect the expression of MMP-2 and MMP-9 proteins.The mRNA expressions of invasion and migration related genes MMP-2 and MMP-9 in each group were detected by qRT-PCR.Results:Berberine hydrochloride could significantly inhibit the proliferation of CAL-27 cells and SCC-15 cells in a concentration and time dependent manner.Berberine hydrochloride can significantly inhibit the migration and invasion of oral squamous cell carcinoma cells.The protein and mRNA expression levels of MMP-2 and MMP-9 were significantly decreased, and the differences were statistically significant(P<0.05).Conclusion:Berberine hydrochloride can effectively inhibit the proliferation, migration and invasion of human oral squamous cell carcinoma cells, and it may play an anti-tumor role by regulating the expression of MMP-2 and MMP-9.
