子宫内膜癌中细胞凋亡和相关基因的表达研究 被引量:6

Apoptosis and related gene expression in endometrial cancer

摘要 目的探讨子宫内膜癌中细胞凋亡和B淋巴细胞瘤-2基因(bcl-2)、BCL2-Associated X蛋白(bax)、天冬氨酸特异性半胱氨酸蛋白酶-3(caspase3)基因的表达研究。方法选取2018年2月至2020年6月间空军军医大学附属西京医院收治的60例子宫内膜癌患者、50例子宫肌瘤患者和30例正常子宫内膜增生患者手术切除的子宫组织。采用脱氧核糖核苷酸末端转移酶介导的缺口末端标记法(TUNEL)检测样本中细胞凋亡率,采用免疫组化染色SP法分别检测样本中bcl-2、bax和caspase3基因的表达水平。结果子宫内膜癌的细胞凋亡率低于子宫肌瘤和正常子宫内膜增生,差异均有统计学意义(均P<0.05)。bcl-2的阳性表达高于子宫肌瘤和正常子宫内膜增生,差异均有统计学意义(均P<0.05)。bax和caspase3的阳性表达率低于子宫肌瘤和正常子宫内膜增生,差异均有统计学意义(均P<0.05)。子宫内膜癌bcl-2、bax和caspase3阳性表达在不同分化程度方面比较,差异均有统计学意义(均P<0.05)。bcl-2、bax和caspase3阳性表达在不同临床分期方面比较,差异无统计学意义(P>0.05)。结论子宫内膜癌组织中细胞凋亡率较低,与bax和caspase3基因表达呈正相关,与bcl-2基因表达呈负相关,bcl-2、bax和caspase3基因与子宫内膜癌的分化程度关系密切,与子宫内膜癌的临床分期无明显相关性。 Objective To investigate apoptosis and expression of B-cell lymphoma-2 gene(Bcl-2),Bcl2-associated X protein(Bax)and aspartate-specific cysteine protease-3(caspase-3)gene in endometrial cancer.Methods From February 2018 to June 2020,60 patients with endometrial cancer,50 patients with uterine fibroids and 30 patients with normal uterine hyperplasia were selected.Uterine tissues were removed surgically.Apoptosis rate was detected in samples by deoxyribonucleotide terminal transferase-mediated nick end labeling(TUNEL),and expression of Bcl-2,Bax and caspase-3 genes were detected by immunohistochemical staining SP method.Results The apoptotic rate was significantly lower in endometrial cancer than in uterine myoma and normal uterine hyperplasia(all P<0.05).The positive expression of Bcl-2 was significantly higher in endometrial cancer than in uterine myoma and normal uterine hyperplasia(all P<0.05).The positive expression rates of Bax and caspase 3 were significantly lower in endometrial cancer than in uterine myoma and normal uterine hyperplasia(all P<0.05).The differences in the positive expression of Bcl-2,Bax and caspase-3 were statistically significant in endometrial cancer of different differentiation degrees(all P<0.05).However,there was no significant difference in the positive expression of Bcl-2,Bax and caspase-3 in different clinical stages(P>0.05).Conclusion Apoptosis rate is low in endometrial cancer tissues,which was positively associated with Bax and caspase-3 gene expression and negatively associated with Bcl-2 gene expression.Bcl-2,Bax and caspase-3 genes are closely associated with differentiation degree of endometrial cancer,but not with clinical stage of endometrial cancer.
