连术消渴颗粒对2型糖尿病大鼠表观指标的影响 被引量:2

摘要 目的:观察连术消渴颗粒对2型糖尿病大鼠模型血糖、食量、饮水量、体重的影响。方法:选取体质量200~220 g的8周龄雄性SD大鼠60只,通过高脂高糖饲料喂养联合腹腔注射链脲佐菌素(STZ 45 mg/kg)诱导大鼠形成2型糖尿病模型,将造模成功的50只大鼠采用随机数字表法分为高剂量组(11.68 g·kg^(-1)·d^(-1))、中剂量组(5.84 g·kg^(-1)·d^(-1))、低剂量组(2.92 g·kg^(-1)·d^(-1))、二甲双胍组(0.17 g·kg^(-1)·d^(-1))、模型组。另取10只健康小鼠作为空白组。每天给药1次,连续给药30天。检测各组大鼠饮食量、饮水量、体重的变化,检测各组大鼠血清空腹血糖(FBG)、糖基化血清蛋白(GSP)水平。结果:连术消渴颗粒能够改善大鼠糖尿病表观指标,降低大鼠饮食量、饮水量,差异具有统计学意义(P﹤0.05);二甲双胍组、连术消渴颗粒高、中、低剂量组可延缓大鼠体重下降趋势,且显著高于模型组(P﹤0.01);与模型组比较,连术消渴颗粒高剂量组、中剂量组给药后,可使大鼠血清FBG、GSP水平显著下降(P﹤0.01);连术消渴颗粒低剂量组可使大鼠血清中GSP水平显著下降(P﹤0.01),FBG水平明显降低(P﹤0.05)。结论:连术消渴颗粒可以降低STZ合并特殊膳食致2型糖尿病大鼠模型的血糖水平、改善饮食量、饮水量、体重等指标,对2型糖尿病具有一定的防治作用。 Objective:To approach the effect of Lianzhu Xiaoke granule on blood glucose,food intake,water intake and body weight of rat model with type 2 diabetes.Methods:60 male SD rats of 8 weeks of age and with 200~220 g body mass were selected.The rats were induced to form the model of type 2 diabetes by feeding with high fat and sugar diet and intraperitoneal injection of streptozotocin(STZ 45mg/kg).By random number table method,50 successfully modeled rats were divided into high-dose group(11.68 g·kg^(-1)·d^(-1)),medium-dose group(5.84 g·kg^(-1)·d^(-1)),low-dose group(2.92 g·kg^(-1)·d^(-1)),metformin group(0.17 g·kg^(-1)·d^(-1))and model group.Other 10 healthy mice were selected as blank group.The drug was administered once a day,continuously for 30 days.The changes of diet intake,water intake and body weight of rats in each group were detected,and the levels of fasting blood glucose(FBG)and glycolated serum protein(GSP)in serum of rat in each group were detected.Results:Lianzhu Xiaoke granule could improve the apparent indices of diabetes in rats,reduce the diet intake and water intake of rats,and the difference was statistically significant(P﹤0.05).Metformin group and Lianzhu Xiaoke granule high-,medium-and low-dose groups could delay the trend of body-weight loss of rats,which was significantly better than model group(P﹤0.01).Compared with model group,Lianzhu Xiaoke granule high-and medium-dose groups could significantly reduce the levels of FBG and GSP in serum of rats after administration(P﹤0.01).Lianzhu Xiaoke granule low-dose group could significantly reduce the level of GSP in serum of rats(P﹤0.01),and the level of FBG was also obviously reduced(P﹤0.05).Conclusion:Lianzhu Xiaoke granule can reduce the level of blood glucose in rat model with type 2 diabetes caused by STZ and special diet,improve the food intake,water intake and body weight etc.apparent indices,which has a certain role for prevention and treatment of type 2 diabetes.
