玉屏风多糖脂质体在雏鸡小肠的吸收特性 被引量:1

摘要 为了研究玉屏风多糖脂质体(YPF-PL)在雏鸡小肠的吸收特性,取12只45日龄SPF雏鸡,建立雏鸡在体小肠循环灌流模型,采用柱前衍生化HPLC分析方法,检测循环回路中不同时间点的药物含量,分别考察雏鸡小肠全肠段、十二指肠、空肠和回肠对YPF-PL和玉屏风多糖(YPF-P)灌流4 h的吸收情况,并通过计算比较药物的累计吸收量及吸收动力学参数:吸收速率常数(Ka)、吸收转化率(A)、吸收半衰期(T_(1/2))及表观渗透系数(P_(app))。结果显示,在小肠全肠段考察中,雏鸡对YPF-PL的吸收总量显著高于传统剂型的YPF-P(P<0.05);雏鸡小肠对YPF-PL的吸收速率常数、转化率和表观渗透系数比YPF-P分别提高了35%,26%,20%,且均显著高于YPF-P(P<0.05);雏鸡小肠对YPF-PL的吸收半衰期比YPF-P缩短了26%,且显著低于YPF-P(P<0.05);在不同肠段考察中,YPF-PL和YPF-P吸收速率常数(Ka)和吸收转化率的大小顺序均为空肠>回肠>十二指肠;与YPF-P相比,YPF-PL在十二指肠、空肠、回肠的吸收速率常数(Ka)分别提高了17%,28%,12%,吸收转化率分别提高了15%,23%,19%,且均极显著高于YPF-P(P<0.01)。结果表明,YPF-PL和YPF-P在雏鸡十二指肠、空肠和回肠都有不同程度的吸收,吸收速率和单位吸收量大小顺序均为空肠>回肠>十二指肠,且与YPF-P相比YPF-PL能提高雏鸡小肠对YPF-P的吸收速率和单位吸收量,进而提高生物利用度,增强疗效。 In order to study the absorption characteristics of Yupingfeng polysaccharide liposome(YPF-PL)in the small intestine of chicks,12 SPF chickens aged 45 d were selected to establish the intestinal circulation perfusion model in vivo,pre-column derivatization HPLC method was used to determine the drug content in the circulation circuit at different time points.The absorption of YPF-PL and YPF-P in the whole intestine,duodenum,jejunum and ileum of chicks was investigated,respectively.The cumulative absorption and absorption kinetic parameters of YPF-PL,the absorption rate constant(Ka),absorption conversion rate(A)and absorption half-life(T_(1/2))were compared by calculation T_(1/2) and apparent permeability coefficient(Papp).The results showed that the total absorption of YPF-PL in chicks was significantly higher than that of YPF-P(P<0.05),and the absorption rate constant,conversion rate and apparent permeability coefficient of YPF-PL in small intestine of chicks were 35%,26%and 20%higher than those of YPF-P(P<0.05);the absorption half-life of YPF-PL was 26%shorter than that of YPF-P and was significantly lower than that of YPF-P(P<0.05);the order of absorption rate constant(Ka)and absorption conversion rate of YPF-PL and YPF-P was jejunum>ileum>duodenum;compared with YPF-P,the absorption rate constant(Ka)of YPF-PL in duodenum,jejunum and ileum increased by 17%,28%and 12%,respectively,and the absorption conversion rate increased by 15%,23%and 19%,respectively,which were significantly higher than YPF-P(P<0.01).These results indicate that YPF-PL and YPF-P has different degrees of absorption in duodenum,jejunum and ileum,and the absorption rate and unit absorption amount is in the order of jejunum>ileum>duodenum,YPF-PL can increase the absorption rate and unit absorption amount of YPF-P in small intestine of chicks compared with YPF-P,thus improving the bioavailability and enhancing the curative effect.
