摘要
目的:分析太白通络凝胶膏对膝骨关节炎兔软骨细胞凋亡的抑制作用及炎性因子表达水平的影响,并探讨其作用机理。方法:采用改良Hulth法制备膝骨关节炎兔模型,ELISA检测兔关节液IL-6、IL-1β、TNF-α含量变化情况;免疫组化检测关节软骨中MMP-13、TGF-β1、Col-Ⅱ表达水平;取兔膝关节软骨制作石蜡切片,并用苏木精-伊红(HE)染色、甲苯胺蓝(TB)染色,用改良Mankin’s标准进行评分。结果:太白通络凝胶膏可降低兔膝关节液中炎症因子IL-6、IL-1β、TNF-α表达水平,差异有统计学意义(P<0.05)。太白通络凝胶膏可提高兔膝关节软骨组织中TGF-β1、Col-Ⅱ的表达水平,降低MMP-13表达水平,差异有统计学意义(P<0.05)。结论:太白通络凝胶膏能降低兔膝关节液及关节软骨中IL-6、IL-1β、TNF-α、MMP-13的表达,促进TGF-β1、Col-Ⅱ的表达,缓解膝关节软骨细胞的凋亡,从而对关节软骨起到保护及修复的作用。
Objective:To analyze the efficacy of Taibai Tongluo gel cream on the inhibition of chondrocyte apoptosis and the expression level of inflammatory factors in rabbits with knee osteoarthritis,and to explore its mechanism of action.Methods:Rabbit model of knee osteoarthritis was prepared by the modified Hulth method,and the changes of IL-6,IL-1βand TNF-αcontents in rabbit joint fluid were detected by ELISA.The expression levels of MMP-13,TGF-β1 and Col-Ⅱin articular cartilage were detected by immunohistochemical detection.Paraffin sections were made from rabbit knee cartilage and stained with hematoxylin-eosin(HE)and toluidine blue(TB),and was scored with modified Mankin’s criteria.Results:Taibai Tongluo gel cream reduced the expression levels of inflammatory factors IL-6,IL-1βand TNF-αin rabbit knee joints,and the difference was statistically significant(P<0.05).Taibai Tongluo gel cream increased the expression levels of TGF-β1 and Col-Ⅱ and decreased the expression level of MMP-13 in rabbit knee cartilage tissue,and the difference was statistically significant(P<0.05).Conclusion:Taibai Tongluo gel cream can reduce the expression of IL-6,IL-1β,TNF-αand MMP-13 in rabbit knee arthritis cartilage,promote the expression of TGF-β1 and Col-Ⅱ,and alleviate the apoptosis of knee joint chondrocytes,thus playing aprotective and repairing role in joint cartilage.
