摘要
通过探究裕民贝母在生理休眠解除过程中营养物质的变化情况,以期揭示裕民贝母种子生理破眠的响应机制。本研究以完成形态休眠的裕民贝母种子为试材,设置CK(蒸馏水对照组)和GA350 mg/L两种处理,于人工智能气候箱5℃~10℃条件下培养,每隔10 d测定营养物质的含量变化。结果发现,随着处理时间的增长,相对含水量增加,细胞的代谢能力增强,种子解除休眠进程加快;粗脂肪和淀粉含量均下降,可能被脂肪酶和淀粉酶分解为小分子化合物供种子萌发使用;可溶性蛋白质和氨基酸含量表现出先升后降,符合种子内部营养物质前期积累后期消耗的特点,营养物质的消耗与生理休眠关系不大,贮藏物质的降解是为了后期的萌发做准备,在GA350 mg/L的处理下,种子内部的生理活动增强,粗脂肪消耗、淀粉消耗均提前,且在氨基酸动员方面也表现出优于CK处理。本研究结果为裕民贝母种子的生产及种质资源的保存提供相应理论依据和技术支持。
To e xplore the changes of nutrients in the process of physiological dormancy breaking of the seed Fritillaria yuminensis X.Z.Duan and to reveal the response mechanism of seed physiological dormancy breaking in Fritillaria yuminensis X.Z.Duan.The experiment used seeds of Fritillaria yuminensis X.Z.Duan which completed the morphological dormancy as experimental materials,were treated with CK(distilled water control group)and GA350 mg/L,were cultured in an artificial intelligence climate box at 5℃~10℃.The nutritiong changes were measured every 10 days.The results showed that,with the growth of the processing time,relative water content increased,the metabolic ability of cells increased,the process of releasing dormancy of seeds accelerated;the content of crude fat and starch decreased,which might be decomposed by lipase and amylase into small molecular compounds for seed germination;the content of soluble protein and amino acid increased at first and then decreased,which accorded with the characteristics of the accumulation and consumption of nutrients in seeds,the consumption of nutrients had little relationship with physiological dormancy,and the degradation of storage substances was for the preparation of g ermination.Meanwhile,GA350 mg/L can enhance the physiological activities of seed,make the changes of physiological more active.The consumption of crude fat and starch are both increased,and it is better than CK in the mobilization of amino acids.The results provide theoretical basis and technical support for seed production and germplasm conservation of Fritillaria yuminensis X.Z.Duan.
