摘要
目的:探讨四次跨膜蛋白1(TM4SF1)在乳腺癌细胞侵袭转移中的作用和可能机制。方法:合成靶向TM4SF1的siRNA并转染MDA-MB-231细胞(TM4SF1 siRNA组),以转染阴性对照siRNA细胞和未转染细胞为对照。用CCK-8实验、Transwell实验检测3组细胞增殖和侵袭转移能力;采用Western blot法检测TM4SF1、E-cadherin、Vimentin、α-SMA、N-cadherin、β-catenin、Snail、Slug、ZEB、TWIST蛋白的表达。结果:与两对照组比较,TM4SF1 siRNA组细胞中TM4SF1蛋白表达降低,增殖、侵袭与转移能力下降(P<0.05);上皮表型E-cadherin蛋白表达增高,而间质表型Vimentin、α-SMA、N-cadherin表达下降(P<0.05);EMT相关转录抑制因子Snail和TWIST表达下降(P<0.05)。结论:TM4SF1可能通过调控乳腺癌细胞EMT进而参与其侵袭转移过程。
Aim:To investigate the role and mechanism of transmembrane-4-L-six-family-1(TM4SF1)in invasion and metastasis of breast cancer cells.Methods:TM4SF1 siRNA was synthesized and transfected into MDA-MB-231 cells(TM4SF1 siRNA group),meanwhile MDA-MB-231 cells transfected with negative siRNA and vacant MDA-MB-231 cells were used as control groups.Next,CCK-8 and Transwell assay were used to detect the proliferation,invasion and metastasis abilities of cells.Western blot was used to detect the expressions of TM4SF1,E-cadherin,Vimentin,α-SMA,N-cadherin,β-catenin,Snail,Slug,ZEB and TWIST.Results:Compared with the two control groups,the expression of TM4SF1 was decreased in cells of TM4SF1 siRNA group,and the proliferation,invasion and metastasis abilities were lower(P<0.05),the expression of E-cadherin(a phenotypic marker of epithelial cells)was higher,the expressions of Vimentin,α-SMA,N-cadherin(the phenotypic markers of mesenchymal cells)were lower(P<0.05),and the expressions of epithelial-mesenchymal transition(EMT)related transcription inhibitors Snail and TWIST were also decreased(P<0.05).Conclusion:TM4SF1 may participate in the invasion and metastasis of breast cells via regulating EMT.
