摘要
目的:研究探讨当归芍药散防治非酒精性脂肪肝(NAFLD)的作用与肠道微生态的相关性。方法:60只SD雄性大鼠随机分为正常组、模型组、阳性药组以及当归芍药散低、中、高剂量组,每组10只。通过喂饲高脂饲料复制NAFLD大鼠模型,造模同时给予相应药物治疗,8周后,采集血清、肝组织和大鼠粪便,检测血清中胆固醇(TC)、甘油三酯(TG)、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)和肝组织中TC、TG及游离脂肪酸(FFA)的含量或活性;苏木精-伊红(HE)染色法观察肝脏病理形态学变化;16sRNA测序法分析粪便肠道菌群组成结构。结果:模型组大鼠肝细胞内出现大量脂滴空泡积聚和气球样变,血清中TC、TG、ALT、AST以及肝组织中TC、TG、FFA的含量或活性均显著升高(P<0.01),Simpon指数、厚壁菌门相对丰度和厚壁菌门/拟杆菌门的数值显著升高(P<0.01),Chao1、Ace、Shannon指数、拟杆菌门相对丰度均显著降低(P<0.01)。与模型组相比,当归芍药散各剂量组大鼠肝细胞内脂滴空泡积聚和气球样病变均明显改善,血清中TC、TG、ALT、AST和肝组织中TC、TG、FFA的含量或活性均明显降低(P<0.05或P<0.01),Chao1、Ace、Shannon指数、拟杆菌门相对丰度均明显升高(P<0.01),Simpon指数、厚壁菌门相对丰度和厚壁菌门/拟杆菌门的数值均显著降低(P<0.01)。相关性分析结果表明,厚壁菌门、拟杆菌门分别与血脂含量呈正、负相关。结论:当归芍药散可能通过改善肠道菌群紊乱进行脂质代谢的调节,进而发挥降脂护肝的作用。
To study the relationship between the effect of Danggui Shaoyao San on prevention and treatment of nonalcoholic fatty liver disease(NAFLD)and intestinal microecology.Methods:60 male SD rats were randomly divided into normal group,model group,positive drug group and low,medium and high dose of Danggui Shaoyao San groups,with 10 rats in each group.NAFLD rat model was established by feeding high-fat diet and given corresponding drug treatment.After 8 weeks,serum,liver tissue and rat feces were collected to detect the contents or activities of cholesterol(TC),triglyceride(TG),alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum the contents of TC,TG and free fatty acid(FFA)in liver tissue;Hematoxylin eosin(HE)staining was used to observe the pathomorphological changes of liver;The composition and structure of fecal intestinal flora were analyzed by 16sRNA sequencing.Results:In the model group,a large number of lipid droplet vacuole accumulation and ballooning degeneration occurred in hepatocytes.The contents or activities of TC,TG,ALT and AST in serum and TC,TG and FFA in liver tissue increased significantly(P<0.01),Simpon index,relative abundance of Firmicutes and values of Firmicutes/Bacteroidetes increased significantly(P<0.01),Chao1,ACE,Shannon index and the relative abundance of Bacteroidetes decreased significantly(P<0.01).Compared with the model group,the lipid droplet vacuole accumulation and ballooning degeneration in hepatocytes in each Danggui Shaoyao San group were significantly improved,the contents or activities of TC,TG,alt,AST in serum and TC,TG and FFA in liver tissue were significantly deceased(P<0.05 or P<0.01),the Chao1,ACE,Shannon indexes and the relative abundance of Bacteroidetes were significantly increased(P<0.01),Simpon index,the relative abundance of Firmicutes and the values of Firmicutes/Bacteroidetes decreased significantly(P<0.01).The results of correlation analysis showed that Firmicutes and Bacteroidetes were positively and negatively correlated with blood lipid co
