基于16SrDNA测序技术揭示腐殖酸对浸矿微生物菌群结构变化的影响

Effect of Humic Acid on the Change of Microbial Community Structure in Ore Leaching Revealed by 16SrDNA Sequencing Technology

摘要 本文利用腐植酸研究了有机碳和空气对浸矿微生物生长曲线和菌群结构变化的影响。菌密度、pH、氧化还原电位(Eh)和Fe^(2+)浓度用于监测微生物生长曲线。高通量测试技术(16SrDNA)被用来研究浸矿微生物组成及变化特点。结果表明,腐植酸的加入导致浸矿微生物以游离菌和吸附菌两种形式存在。腐植酸和空气的协同作用能显著加快细菌的生长繁殖。在含有腐植酸的细菌生长环境中,其菌密度比不含腐植酸的菌密度高29.59%。密封环境(无空气)导致菌密度最终低于10^(8) cells/mL,即腐植酸和空气的协同强化效果比单一条件显著。高通量测试结果表明,腐植酸和空气的协同作用主要是促进培养液中吸附细菌菌属的生长和繁殖。此外,腐植酸的存在导致浸矿微生物菌群中出现了可固定分解有机碳的菌属,包括叶瘤杆菌属(Phyllobacterium)、慢生根瘤菌属(Bradyrhizobium)、中慢生根瘤菌属(Mesorhizobium)、鞘氨醇单胞菌属(Sphingomonas)和类杆菌属(Bacteroides)。 Humic acid is used to study the effects of organic carbon and air on the growth curve of leaching microorganisms and the changes in the microbial community structure in this paper.Bacterial density,pH,redox potential(Eh)and Fe^(2+)concentration are used to monitor the growth curve of microorganisms.High-throughput testing technology(16SrDNA)is used to study the composition and change characteristics of leaching microorganisms.The results show that the addition of humic acid causes the leaching microorganisms to exist in two forms:free bacteria and adsorbed bacteria.The synergistic effect of humic acid and air can significantly accelerate the growth and reproduction of bacteria.In the growth environment of bacteria containing humic acid,the density of bacteria is 29.59%higher than that of bacteria without humic acid.The sealed environment(no air)results in the final bacterial density being lower than 10^(8)cells/mL,that is,the synergistic strengthening effect of humic acid and air is more significant than that of a single condition.High-throughput test results show that the synergistic effect of humic acid and air is mainly to promote the growth and reproduction of adsorbed bacteria in the culture solution.In addition,the presence of humic acid leads to the emergence of bacteria that can fix and decompose organic carbon in the leaching microbial flora,including Phyllobacterium,Bradyrhizobium,Mesorhizobium,Sphingomonas and Bacteroides.
