摘要
于安徽省池州市石台大山村富硒土壤中分离得到富硒菌株,对其进行分类鉴定和耐硒能力的研究及培养条件的优化.根据土壤梯度稀释法分离出几种纯菌株,选取优势菌株经形态观察和16SrRNA鉴定其分类地位.研究培养基硒浓度对该菌生长的影响,通过温度和时间优化培养条件.获得一富硒菌株C5,革兰染色阳性,经16SrRNA鉴定为假真菌样芽孢杆菌;该菌株在硒浓度15-30 g/ml培养基中生长良好,最佳浓度为25 g/ml;C5菌在28-35℃和培养24-96 h均能生长良好,优化后C5菌培养的最佳条件是温度32℃,培养时间72 h.C5菌耐硒能力较好,对温度和培养时间的要求不是非常苛刻,具有研究和应用价值.
A selenium-enriched strain was isolated from high-Se area in Dashan Village,Shitai,Chizhou City,Anhui Province.Then the strain was identified and its selenium tolerance and cultivation conditions were studied.Several strains were isolated with the method of gradient dilution of soil.The dominant strain was identified according to morphological observation and 16SrRNA.And it was cultivated on various conditions of selenium concentration in the optimization of culture conditions by temperature and time.The strain which was gram positive named as C5 was identified as Bacillus pseudomycoides by 16SrRNA.It grew well in the medium with selenium concentration of 15-30g/ml at 28-35℃and 24-96h and the optimum concentration was 25g/ml.The optimum conditions for the growth of the strain were:temperature 32℃,culture time 72h.C5 which was good at selenium tolerance had no strict temperature and culture time requirements.This study provided a good strain for the further studies.
