Determination of Five Alternaria Toxins in Tomato Ketchup Using UPLC-MS/MS

摘要 该试验建立了超高效液相色谱-串联质谱(UPLC-MS/MS)测定番茄酱中5种链格孢菌毒素(腾毒素、链格孢霉素、交链格孢酚单甲醚、链格孢酚及细交链格孢菌酮酸)的测定方法。番茄酱样品经乙腈提取, QuEChERS方法净化;采用C18色谱柱分离,以0.1%甲酸水和乙腈作为流动相进行梯度洗脱。以电喷雾离子源、正离子扫描和多反应监测(MRM)模式检测,外标法定量。结果表明, 5种链格孢霉毒素在0.5~200 ng/mL质量浓度范围内线性关系良好,相关系数均大于0.99。5种链格孢霉毒素的方法检出限均为1μg/kg,方法定量限均为2.5μg/kg;平均回收率在78.3%~104.8%之间,相对标准偏差为1.2%~7.4%(n=6)。该方法简单、灵敏度高、分析时间短,适用于番茄酱中5种链格孢霉毒素的测定。 A method was established for determining five Alternaria toxins(tentoxin, alternaria, alternariol monomethylether, alternariol and tenuazonic acid) in tomato ketchup by ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS). Samples were extracted with acetonitrile, and purified by the QuEChERS method. Five Alternaria toxins(ATs) were separated on a BEH C18 column with gradient elution using a mobile phase made up of 0.1% formic water and acetonitrile. The samples were detected under positive multiple reactions monitoring(MRM). The results were quantified by external standard method. The linearities of five ATs were in the concentration range of 0.5-200 ng/mL, with the correlation coefficients higher than 0.99. The limits of detection(LOD) and the limits of quantitation(LOQ) of the method were1 μg/kg and 2.5 μg/kg, respectively. The average spiked recovery rates were in the range of 78.3%-104.8%. The relative standard deviation(SRSD, n=6) was 1.2%-7.4%. The method proved to be simple, sensitive, rapid and suitable for the determination of five ATs in tomato ketchup.
