摘要
为研究猪不同品种的MYL1基因的多态性及mRNA表达差异。本试验以藏猪和大约克猪作为实验动物,采用一代测序技术对藏猪(59头)和大约克猪(58头)MYL1基因起始密码子上游3 kb区域进行多态性检测;利用实时荧光定量PCR技术检测MYL1基因在背最长肌、背脂、肝脏组织差异表达情况。结果表明:藏猪和大约克猪在起始密码子上游3 kb区域发现T-159A突变位点并呈极显著差异(P<0.01);MYL1基因在背最长肌表达量最高,其次是背脂,肝脏的最低,在背最长肌中藏猪MYL1基因表达量极显著高于大约克猪(P<0.01),在背脂和肝脏组织中,藏猪MYL1基因表达量显著高于大约克猪(P<0.05)。推测T-159A位点可能是参与肌肉生长发育的重要调控位点。本研究为下一步开展MYL1基因对猪生长性能及其肉品质的影响提供参考依据。
In order to study the polymorphisms and mRNA expression differences of different breeds of pig MYL1 gene,in this experiment,Tibetan pigs and Yorkshire pigs were used as experimental animals.Polymorphism detection was performed on the 3 kb region upstream of the start codon of the MYL1 gene in Tibetan pigs(59 heads)and Yorkshire pigs(58 heads)by one-generation sequencing technology.Differential expression of MYL1 gene in longissimus dorsi muscle,back fat,and liver tissue was detected by RT-qPCR.The results showed that the T-159 A mutation site was found in the 3 kb region upstream of the start codon of Tibetan pigs and Yorkshire pigs and showed a significant difference(P<0.01);The expression of MYL1 gene was the highest in the longest muscle of the back,followed by back fat,the liver is the lowest,the expression of MYL1 gene in the longest muscles of MYL1 gene was significantly higher than that of Yorkshire pigs(P<0.01),and the expression of Tibetan pigs in back fat and liver tissues of Tibetan pigs was significantly higher than that of Yorkshire pigs(P<0.05).It is speculated that the T-159 A site may be an important regulatory site involved in muscle growth and development.This study provides a reference for the next step in the development of the effect of the MYL1 gene on pig growth performance and meat quality.
