摘要 目的:通过生物信息学方法分析脂多糖结合蛋白(LBP)在胃癌组织中的表达及意义。方法:从Ualcan数据库获取正常胃组织及胃癌组织LBP表达数据,利用GEPIA数据库分析LBP表达与胃癌患者预后的关系,在LinkedOmics中检索LBP在胃癌中的共表达基因,通过GO富集分析、KEGG通路分析确定这些基因参与的生物学过程及功能。最后利用STRING数据库分析相关基因编码蛋白的互作网络。结果:与正常胃组织相比,Ⅰ和Ⅲ分期、老年和男性患者胃癌组织中LBP表达水平均明显升高,但与幽门螺杆菌感染与否无显著相关性。在胃癌中与LBP共表达的基因主要涉及急性炎症反应、激活蛋白级联、蛋白质-脂质复合物亚基组织等生物学过程,介导货物受体活性、脂多糖结合、蛋白质-脂质复合物的绑定、糖胺聚糖绑定等分子功能,富集于血液微粒、蛋白质-脂质复合物、内质网腔等细胞组分。富集的KEGG信号通路主要与补体和凝血级联、胆固醇代谢、PPAR信号等通路相关。LBP高表达患者的总生存率和无病生存率明显低于LBP低表达患者。LBP蛋白与TLR4、STAT3、IL-6、STAT6、IL-4、IL-10、JAK1、JAK2、JAK3、SOCS3等存在互作关系。结论:LBP在胃癌组织中高表达,且与胃癌的发生、发展及预后密切相关,可以作为胃癌筛选、预后预测的标志物之一。 Objective:To analyze expression levels of LBP in gastric cancer,explore its clinical significance during gastric cancer development by bioinformatics analysis.Methods:Using Ualcan gastric cancer data to test the expression difference in gastric tumor tissues and normal tissues. Relationship between expression of LBP and prognosis of gastric cancer patients was studied by GEPIA. Co-expressed genes of LBP were retrieved in LinkedOmics database,and the biological processes and functions of these genes were determined by GO enrichment analysis and KEGG pathway analysis. Finally,the interaction network of proteins encoded by relevant genes was analyzed by STRING database.Results:Compared with normal tissue,expression levels of LBP in gastric cancer tissues from patients withⅠand Ⅲ disease stages,elderly and male patients were significantly increased. Expression of LBP had no significant correlation with H.pylori infection. Co-expressed genes of LBP mainly involved biological processes such asacute inflammatory response,protein activation cascade and protein-lipid complex subunit organization,and mediated molecular functions such as cargo receptor activity,lipopolysaccharide binding and protein-lipid complex binding,and were enriched in blood microparticle,protein-lipid complex and endoplasmic reticulum. Enriched pathways were primarily involved in complement and coagulation cascades,cholesterol metabolism and PPAR signaling pathway. The overall survival rates and disease-free survival rates in gastric cancer patients with high LBP expression were significantly lower than those patients with low LBP expression. TLR4,STAT3,IL-6,STAT6,ITGAM,IL-10,IL-4,SRC,EGFR,JAK1,AKT1 and CD14 were found having interactions with LBP protein.Conclusion:LBP is highly expressed in gastric cancer tissues,and its expression level has closely associated with development of gastric cancer.
