摘要
目的研究创伤后应激障碍(posttraumatic stress disorder,PTSD)小鼠海马神经细胞内FoxO3a和LC3蛋白表达的变化。方法采用单一连续刺激(single prolonged stress,SPS)方法建立PTSD小鼠模型,应用旷场实验、高架十字迷宫、Morris水迷宫3种行为学检测方法测试SPS刺激后小鼠的空间记忆能力和焦虑水平,通过Western blot法检测海马神经细胞自噬相关蛋白LC3、p62以及信号通路相关蛋白Akt、p-Akt、FoxO3a、p-FoxO3a的表达,并利用免疫荧光染色方法观察FoxO3a、LC3的表达水平。结果行为学实验检测结果显示,与Control组比较,PTSD组小鼠焦虑水平升高及空间记忆受损(P<0.01);Western blot法和免疫荧光染色结果显示,PTSD组小鼠海马神经细胞p-Akt蛋白水平增加,FoxO3a蛋白表达增加,p-FoxO3a蛋白表达减少,自噬相关蛋白LC3Ⅱ/LC3Ⅰ比值增加,p62蛋白水平降低(P<0.05);海马神经细胞内FoxO3a和LC3荧光表达强度增加。结论SPS刺激后PTSD小鼠海马神经细胞自噬增多,FoxO3a表达增加。
Objective To study the expression changes of FoxO3a and LC3 protein in hippocampal nerve cells of mice with posttraumatic stress disorder(PTSD).Methods The PTSD mice model was established by single prolonged stress(SPS).tThe Open Field Test,the Elevated plus maze and the Morris water maze were used to detect the spatial memory ability and anxiety level of mice after SPS.The levels of hippocampal nerve cells autophagy related proteins LC3,p62 and signaling pathway related proteins Akt,p-Akt,FoxO3a,p-FoxO3a were detected by Western blotting,and the expression levels of FoxO3a and LC3 were observed by immunofluorescence staining.Results The results of behavioral experiments showed that the anxiety level and impaired spatial memory in PTSD group were significantly higher than in Control group(P<0.01).The results of Western blot and immunofluorescence staining showed that The levels of p-Akt,FoxO3a protein and autophagy related protein LC3II/LC3I in hippocampal nerve cells were increased in PTSD group than in Control group,and the levels of p-FoxO3a,p62 protein were decreased in PTSD group than in Control group(P<0.05).The immunofluorescence intensity of FoxO3a and LC3 were increased in hippocampal nerve cells.Conclusion Autophagy and FOXO3a expression increased in hippocampal nerve cells of PTSD mice after single prolonged stress(SPS).
