摘要
目的:通过网络药理学方法探讨野菊花治疗高血压病的作用机制。方法:本研究运用中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)结合筛选条件确定野菊花的主要有效活性成分及其作用靶点,构建主要有效活性成分-靶点网络图。利用GeneCards和OMIM数据库获取高血压病靶点,取野菊花和高血压病的交集靶点,对交集靶点进行蛋白质互作网络分析、生物通路及富集分析。结果:共收集到野菊花8个有效成分及其290个成分靶点;高血压疾病靶点7811个;药物成分和疾病的共有靶点170个;最终得到关键核心靶点有蛋白激酶B1(AKT1)、丝裂原活化蛋白激酶1(MAPK1)、RELA、90kDa热休克蛋白αA1(HSP90αA1)、白细胞介素6(Interleukin-6,IL-6)。生物过程(Gene Ontology,GO)功能富集分析得到共2312条GO生物学过程(Biological Process,BP),90个分子功能(Molecular Function,MF)条目,177个细胞组分(Cellular Components,CC)相关的条目。京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)富集分析主要有:糖尿病并发症中的晚期糖基化终产物及其受体(AGE-RAGE)信号通路、磷脂酰肌醇-3-激酶(PI3k)-akt信号通路、流体剪切应力与动脉粥样硬化、IL-17信号通路、肿瘤坏死因子信号通路等。结论:野菊花治疗高血压病具有多成分、多靶点、多通路的特点,其机制较为复杂,本研究结果可为探析中医治疗高血压病提供参考。
Objective:To explore the action mechanism of flos chrysanthemi indici on hypertension based on network pharmacology.Methods:In this study,TCMSP database and screening conditions were used to determine the main active components and their targets of flos chrysanthemi indici,and the main active components target network diagram was constructed.Hypertension targets were obtained through GeneCards and OMIM database,the intersection targets of flos chrysanthemi indici and hypertension were selected,and protein interaction network analysis,biological pathway and enrichment analysis of the intersection targets were carried out.Results:A total of 8 active components and 290 component targets of flos chrysanthemi indici were collected,7811 targets for hypertension,170 targets for medicine components and diseases;finally,the key core targets such as AKT1,MAPK1,RELA,HSP90AA1,IL6 were obtained.A total of 2312 BP,90 MF items and 177 CC related items were obtained by GO functional enrichment analysis.KEGG enrichment analysis mainly includes AGERAGE signaling pathway,PI3K-akt signaling pathway,fluid shear stress and atherosclerosis,IL-17 signaling pathway,tumor necrosis factor signaling pathway and so on.Conclusion:Flos chrysanthemi indici on hypertension is characterized by multi-component,multi-target and multi-channel,and its mechanism is more complex.The results of the study can provide more reference for treating hypertension in TCM.
