摘要
目的观察心肌梗死(MI)大鼠心肌微小RNA(miR)-130a-3p的表达及其对心肌炎症的影响。方法使用随机数字法将雄性SD大鼠随机分为假手术组(Sham组)、MI组、miR-130a-3p组(心肌注射miR-130a-3p mimics)和阴性对照组(心肌注射miR-130a-3p mimics阴性对照),每组15只。心脏彩超测定左心室收缩末期内径(LVDs)、左心室舒张末期内径(LVDd)、左心室长轴缩短分数(FS)、左心室射血分数(LVEF);原位缺口末端标记法(TUNEL)法测定心肌凋亡指数;酶联免疫吸附试验(ELISA)检测血清肌钙蛋白T(TnT)、肌钙蛋白I(TnI)、肌酸激酶同工酶(CK-MB)、N末端B型利钠肽前体(NT-proBNP)、肿瘤坏死因子-α(TNF-α)和白细胞介素(IL)-1β水平;反转录-聚合酶链反应(RT-PCR)法测定心肌组织miR-130a-3p、NOD样受体家族3炎症小体(NLRP3)的表达;蛋白质印迹法(Western blot)测定心肌组织B细胞淋巴瘤/白血病-2相关X蛋白(bax)、B细胞淋巴瘤/白血病-2(bcl-2)和NLRP3的表达水平。各组间比较采用方差分析。结果MI组miR-130a-3p水平明显低于Sham组(t=4.365,P<0.05)。miR-130a-3p组大鼠LVDs和LVDd低于MI组(t=4.475、5.172、P<0.05),FS和LVEF高于MI组(t=3.679、5.442,P<0.05)。miR-130a-3p组血清TnT、TnI、CK-MB、NT-proBNP、TNF-α、IL-1β水平明显低于MI组(t=4.265、5.382、3.976、4.436、7.246、4.854,P<0.05)。Sham、MI、阴性对照组和miR-130a-3p组凋亡指数依次为(5.14±0.62)%、(34.54±4.37)%、(35.27±4.55)%和(18.76±2.48)%,miR-130a-3p组凋亡指数低于MI组(t=4.356,P<0.05)。miR-130a-3p组NLRP3 mRNA和bax蛋白、NLRP3蛋白相对表达量均低于MI组(t=6.296、11.356、8.235,P<0.05),bcl-2蛋白水平相对表达量高于MI组(t=8.876,P<0.05)。结论miR-130a-3p在MI大鼠心肌组织中呈低表达,上调miR-130a-3p表达可以减轻MI大鼠心肌炎症和细胞凋亡。
Objective To study the expression of microRNA(miR)-130a-3p and its effect on myocardial inflammation in rats with myocardial infarction(MI).Methods Male Sprague-Dawley(SD)rats were randomly divided into sham group,MI group,miR-130a-3p group(myocardial injection of miR-130a-3p mimics)and negative control group(myocardial injection of miR-130a-3p NC),15 rats in each group.Left ventricular end systolic diameter(LVDS),left ventricular end diastolic diameter(LVDD),left ventricular long axis shortening fraction(FS)and left ventricular ejection fraction(LVEF)were measured by color Doppler ultrasound.TdT-mediated dUTP-biotin nick end labeling(TUNEL)was used to measure myocardial apoptotic index.Serum troponin T(TnT),troponin I(TnI),creatine kinase-MB(CK-MB),N-terminal fragment of the BNP precursor(NT-proBNP),tumor necrosis factor-α(TNF-α),and interleukin(IL)-1βlevels were detected by enzyme linked immunosorbent assay(ELISA).The expression levels of miR-130a-3p and NLR family,NOD-like receptor family,pyrin domain containing 3(NLRP3)were detected by reverse transcription-polymerase chain reaction(RT-PCR).The expression levels of B cell lymphoma/leukemia-2 associated X protein(bax),B cell lymphoma/leukemia-2(bcl-2)and NLRP3 were determined by Western blotting.Results The level of miR-130a-3p in MI group was significantly lower than that in sham group(t=4.365,P<0.05).As compared with MI group,LVDS and LVDD in miR-130a-3p group were significantly decreased(t=4.475,5.172,P<0.05),FS and LVEF were significantly increased(t=3.679,5.442,P<0.05).Serum TNT,TnI,CK-MB,NT proBNP and TNF-α,IL-1βlevels in miR-130a-3p group were significantly lower than in MI group(t=4.265,5.382,3.976,4.436,7.246,4.854,P<0.05).The apoptotic index in sham group,MI group,negative control group and miR-130a-3p group was(5.14±0.62)%,(34.54±4.37)%,(35.27±4.55)%,and(18.76±2.48)%respectively.The apoptotic index in miR-130a-3p group was significantly lower than that in MI group(t=4.356,P<0.05).As compared with MI group,the level of NLRP3 mRNA,the relativ
