梓醇通过上调BMP-2表达激活Wnt/β-catenin信号通路促进大鼠股骨骨折愈合 被引量:3

摘要 目的:探讨梓醇对大鼠股骨骨折愈合的作用及其可能的作用机制。方法:将48只SD股骨骨折模型大鼠随机分为对照组、梓醇组、梓醇+sh-NC组和梓醇+sh-BMP-2组,每组12只。梓醇组大鼠灌胃50 mg/kg梓醇,梓醇+sh-NC组大鼠灌胃50 mg/kg梓醇和尾静脉注射300μL sh-NC慢病毒液,梓醇+sh-BMP-2组大鼠灌胃50 mg/kg梓醇和尾静脉注射300μL sh-BMP-2慢病毒液,对照组大鼠灌胃等量溶剂,1次/d,连续给药3周。X线检测骨折愈合;Micro-CT检测骨体积(BV)、骨体积分数(BV/TV)、骨小梁数量(TB.N)、骨小梁间距(TB.Sp)和骨小梁厚度(TB.Th);RT-PCR检测BMP-2 mRNA表达;Western blotting检测BMP-2、Runx2、ColⅠ、β-catenin、cyclinD1和c-myc蛋白表达;ELISA检测血清ALP和OCN水平。结果:与对照组比较,梓醇组大鼠X线评分、BV、BV/TV、TB.N、TB.Th、BMP-2 mRNA及蛋白表达显著升高(P<0.05),ALP和OCN水平以及Runx2、ColⅠ、β-catenin、cyclinD1和c-myc蛋白表达显著升高(P<0.05),TB.Sp无显著差异(P>0.05);梓醇组和梓醇+sh-NC组大鼠各项指标差异无统计学意义(P>0.05);与梓醇+sh-NC组相比,梓醇+sh-BMP-2组大鼠X线评分、BV、BV/TV、TB.N、TB.Th、BMP-2 mRNA和蛋白表达显著降低(P<0.05),ALP和OCN水平以及Runx2、ColⅠ、β-catenin、cyclinD1和c-myc蛋白表达显著降低(P<0.05),TB.Sp无显著差异(P>0.05)。结论:梓醇可能通过上调BMP-2表达激活人无翅型MMTV整合位点家族/β-连锁蛋白(Wnt/β-catenin)信号通路促进大鼠股骨骨折愈合。 Objective:To explore the effect of catalpol on rat femoral fracture healing and its possible mechanism.Methods:48 SD femoral fracture model rats were randomly divided into control group,catalpol group,catalpol+sh-NC group and catalpol+sh-BMP-2 group,with 12 rats in each group.Rats in the catalpol group were intragastrically administered with 50 mg/kg catalpol,rats in the catalpol+sh-NC group were given intragastrically with 50 mg/kg catalpol and 300μL sh-NC lentiviral solution via tail vein,rats in catalpol+sh-BMP-2 group were intragastrically administered with 50 mg/kg catalpol and 300μL sh-BMP-2 lentiviral solution via tail vein,and rats in the control group were intragastrically administered with the same amount of solvent,once a day for 3 weeks.X-ray was used to detect fracture healing.Micro-CT was used to detect bone tissue morphology and to measure bone volume(BV),bone volume fraction(BV/TV),bone trabecular number(TB.N),bone trabecular spacing(TB.Sp)and bone trabecular thickness(TB.Th).RT-PCR was used to detect the expression of BMP-2 mRNA.Western blotting was used to detect the expression of BMP-2,Runx2,ColⅠ,β-catenin,cyclinD1 and cmyc protein.ELISA was used to detect the levels of ALP and OCN in serum.Results:Compared with the control group,the X-ray score,BV,BV/TV,TB.N,TB.Th,and expressions of BMP-2 mRNA and protein in the catalpol group were significantly increased(P<0.05),while the levels of ALP and OCN and the expressions of Runx2,ColⅠ,β-catenin,cyclinD1,c-myc protein were significantly increased(P<0.05).There was no significant difference in TB.Sp between the control group and the catalpol group(P>0.05).There was no significant difference in all indicators between the catalpol group and the catalpol+sh-NC group(P>0.05).Compared with the catalpol+sh-NC group,the X-ray score and BV,BV/TV,TB.N,TB.Th,the expressions of BMP-2 mRNA and protein in the catalpol+sh-BMP-2 group were significantly reduced(P<0.05),whereas the levels of ALP and OCN in serum and the expressions of Runx2,ColⅠ,β-catenin,c
