摘要
采用SRAP分子标记技术对18个大蒜群体的遗传多样性进行分析,用Popgene 32软件计算遗传多样性参数,并采用NTSYS pc 21-2软件进行种质间的UPGMA聚类分析和基于遗传一致度的群体间聚类分析。结果表明,22对引物扩增出161条多态性条带,引物的多态性信息指数(PIC)在0.635 7~0.889 7之间。种质间的平均Nei’s遗传多样性指数(H)为0.434 8,平均Shannon’s信息多样性指数(I)为0.623 3,基因分化系数(Gst)为0.360 4,基因流(Nm)为0.887 4。群体间的遗传一致度范围为0.633 5~0.974 4,遗传距离范围为0.025 9~0.456 4,Nei’s基因多样性(H)介于0.144 1~0.355 7之间,Shannon’s信息指数(I)在0.210 3~0.529 7之间。采用NTSYS-sp 2.1软件构建UPGMA聚类图,大蒜种质间和群体间分别在遗传相似系数为0.58和0.83时将供试材料分为五类,其中I类又可分为7个亚类,该聚类结果主要由品种差异决定,受地域影响较小。
In this study, genetic diversity of 18 garlic populations was analyzed by using SRAP molecular markers, and genetic diversity parameters were calculated by Popgene 32 software. And, the NTSYS PC 21-2 software was used to perform UPGMA cluster analysis between germplasms and interpopulation cluster analysis based on genetic consistency. The results showed that 161 polymorphic bands were amplified by 22 pairs of primers, and the polymorphism information index(PIC) of the primers ranged from 0.635 7 to 0.889 7. Additionally, the average Nei’s genetic diversity index(H), Shannon’s information diversity index(I), gene differentiation coefficient(GST) and gene flow(NM) among these garlic germplasms were 0.434 8, 0.623 3, 0.360 4 and 0.887 4, respectively. The genetic consistency between populations ranged from 0.633 5 to 0.974 4, and the genetic distance ranged from 0.025 9 to 0.456 4. And, the Nei’s gene diversity(H) ranged from 0.144 1 to 0.355 7 while the Shannon’s information index(I) ranged from 0.210 3 to 0.529 7. The UPGMA cluster map was constructed by NTSYS-SP 2.1 software, and the 18 garlic germplasms were divided into 5 groups when the genetic similarity coefficient was 0.58 and 0.83, respectively, among which the group I could be further divided into 7 subgroups. The clustering results were mainly determined by the variety differences, but were not affected by the region.
