治疗牛支原体病复方中药提取工艺的优化及其体外抑菌效果 被引量:2

Optimization of extraction process of traditional Chinese medicine for the treatment of Mycoplasma bovine disease and its in vitro bacteriostatic effect

摘要 为优化治疗牛支原体病复方中药提取工艺及研究其体外抑菌效果,为牛支原体病防治及中药制剂的开发提供理论依据,试验采用正交试验对大青叶、黄芩及百部3种中药提取工艺的乙醇浓度、液料比、提取时间及提取温度进行优化。以最佳提取方法得到提取物,按不同比例混合得到中药组方合剂,采用平板打孔法进行体外抑菌试验,并采用二倍稀释法测定其最小抑菌浓度(MIC)和最小杀菌浓度(MBC)。结果表明:大青叶最佳提取工艺为70%乙醇溶液,提取温度70℃,液料比10∶1,提取时间2 h;黄芩最佳提取工艺为60%乙醇溶液,提取温度70℃,液料比40∶1,提取时间1.5 h;百部最佳提取工艺为液料比20∶1,提取温度70℃,50%乙醇溶液,提取时间1.5 h。3种中药提取物按不同比例混合时,抑菌圈平均直径为18.4-22.5 mm,其中大青叶∶黄芩∶百部=2.6∶2.6∶1时对牛支原体体外抑菌效果最好,抑菌圈平均直径为22.5 mm;二倍稀释法测得MIC均为31.25 mg/mL,MBC为31.25-62.50 mg/mL。说明3种中药提取物及不同配比对牛支原体病有明显的抑制作用,可作为中兽药开发利用。 In order to optimize the extraction process of traditional Chinese medicine for the treatment of Mycoplasma bovine disease and study its antibacterial effect in vitro,and to provide theoretical basis for the prevention and treatment of Mycoplasma bovine disease and the development of traditional Chinese medicine preparations,in this study,orthogonal experiment was used to optimize the ethanol concentration,liquid-solid ratio,extraction time and extraction temperature of Folium isatidist,Scutellaria baicalensis Georgi,Stemnae.The extracts were obtained by the best extraction method and mixed in different proportions to obtain the mixture of Chinese medicine.In vitro inhibition test was carried out by plate drilling method,and the minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)were determined by double dilution method.The results showed that the optimal extraction conditions of Folium isatidist were 70%ethanol solution,extraction temperature 70℃,liquid-solid ratio 10∶1,extraction time 2 h.The optimal extraction conditions of Scutellaria baicalensis Georgi were extraction time 1.5 h,60%ethanol solution,extraction temperature 70℃,liquid-solid ratio 40∶1.The optimal extraction conditions of Stemnae were liquid-solid ratio 20∶1,extraction temperature 70℃,50%ethanol solution,extraction time 1.5 h.When the extracts were mixed in different proportions,the average diametres of bacteriostatic rings were 18.4-22.5 mm,among which Folium isatidist∶Scutellaria baicalensis Georgi∶Stemnae=2.6∶2.6∶1 had the best bacteriostatic effect on Mycoplasma bovine in vitro,and the average diametres of bacteriostatic rings were 22.5 mm.The MIC and MBC values were 31.25 mg/mL and 31.25-62.50 mg/mL by double dilution method.The results indicated that the extracts of three traditional Chinese medicines and their different proportions had obvious inhibitory effects on Mycoplasma bovine disease and could be used as Chinese veterinary drugs.
