摘要
番茄褐色皱纹果病毒(tomato brown rugose fruit virus,ToBRFV)是烟草花叶病毒属新成员,番茄和辣椒是其主要寄主,被中国、欧盟、美国、澳大利亚、土耳其、新西兰、智利、阿根廷、俄罗斯、哈萨克斯坦、泰国等国家或地区纳入需检疫的有害生物。我们利用电镜观察、RT-PCR和SYBR Green实时荧光RT-PCR首次从进境的番茄种子检出ToBRFV。电镜观察显示该样品所携带的病毒粒子形态符合烟草花叶病毒属病毒的粒子特征,RT-PCR扩增出特异的条带,实时荧光RT-PCR有明显的扩增曲线,PCR产物测序结果与ToBRFV(GenBank登录号MK133093.1和KT383474.1)同源性高达99.65%。
Tomato brown rugose fruit virus(ToBRFV)is a recently described member of the Tobamovirus genus.Tomato and pepper are the main hosts.To prevent the introduction of ToBRFV,it has been required to quarantine by China,EPPO,the United States,Australia,Turkey,New Zealand,Chile,Argentina,Russia,Kazakhstan and Thailand,etc.We detected and identified ToBRFV from imported tomato seeds for the first time by electron transmission microscope,RT-PCR and SYBR Green real time RT-PCR.The result of observation showed that the virus particle morphology of the sample was consistent with the particle characteristics of Tobamovirus genus.RT-PCR amplied the specific band and SYBR Green real-time RT-PCR had obvious amplification curve and the sequencing results were 99.65%similar to ToBRFV sequence(GenBank login numbers MK133093.1 and KT383474.1).
