摘要
目的建立人胃癌耐药细胞,为研究人胃癌耐药细胞的耐药机制以及为耐药肿瘤细胞的治疗提供耐药肿瘤细胞模型。方法体外培养人胃癌细胞SGC-7901,用50μmol/L顺铂多次反复冲击,对冲击4次和6次的SGC-7901分别检测对顺铂的抑制率和多药耐药基因(MDR)及多药耐药特异基因片段的表达,并计算药物对细胞的半数抑制浓度(IC50)。结果经4次和6次冲击后,受冲击的SGC-7901胃癌细胞对顺铂的抑制率明显高于亲本细胞,IC50为63.98μmol/L和66.34μmol/L,而亲本细胞SGC-7901的IC50则分别为20.55μmol/L和18.22μmol/L,冲击4次和6次的SGC-7901细胞有MDR基因和基因特异产物表达。结论经顺铂反复冲击4次和6次的SGC-7901检测结果证明,SGC-7901对顺铂的耐受性明显增强,其抑制率较对照组高,IC50与对照组相比有明显差异(P<0.01),而且均有耐药基因表达,该细胞可以作为耐药细胞进行相关实验。
Objective To establish drug-resistant human gastric cancer SGC-7901 cells,provide drug resistant cell model to study the drug resistance mechanism of human gastric cancer and the treatment of drug-resistant tumors.Methods Culture human gastric cancer cells SGC-7901 in vitro,repeated lashing them with 50μmol/L cisplatin,detected the inhibition rate and multidrug resistance gene(MDR)expression of SGC-7901 after four and six times cisplatin lashed and calculated the IC50.Results After lashed for four and six times,the inhibitory rate of the SGC-7901 cells have significantly higher than parent cells(P<0.05),IC50 is 63.98μmol/L and 66.34μmol/L separately,while the parent cell SGC-7901 is 20.55μmol/L and 18.22μmol/L correspondingly.The SGC-7901 cells which have been lashed4 and 6 times both have MDR expressions.Conclusion The test results certified that SGC-7901 lashed by cisplatin for four and six times both have significantly enhanced tolerability.The inhibition rate was higher than control group,and the IC50 have significantly difference compared with control group(P<0.01).All of them have resistance gene expression,this results indicated that it can use them as drug-resistant cell for related study.
