摘要
为探究麦冬多糖对鸡新城疫病毒(Newcastle disease virus,NDV)的作用,试验采用水提醇沉法提取5种分子量麦冬多糖,采取三氯乙酸法去除蛋白等杂质,用苯酚-硫酸法和考马斯亮兰G-250法分别测定提取物的糖和蛋白含量,用MTT法测定不同分子量麦冬多糖对鸡胚成纤维细胞(CEF)的安全浓度,用3种作用方式测定麦冬多糖对NDV的作用效果及机制。结果显示:麦冬多糖对CEF的安全浓度为78.125μg/mL;OPS80在3种作用方式中的病毒抑制率分别为37.36%、29.63%、39.30%,均高于其余4种。研究表明,各麦冬多糖均有明显的抗NDV活性,其中OPS80的活性最好,作用机制最可能为杀灭和阻断作用。
In order to explore the anti-Newcastle disease virus(NDV)activity of Ophiopogon japonicus polysaccharides,this experiment used the water extraction and alcohol precipitation method to complete the extraction of 5 kinds of molecular weight Ophiopogon japonicus polysaccharides.and adopted the"trichloroacetic acid method"to remove the proteins.The phenol-sulfuric acid method and Coomassie Brilliant Blue G-250 were used to determine polysaccharide content and protein content,respectively.The safe concentration of polysaccharides of different molecular weight at various levels on chicken embryo fibroblasts(CEF)was determined by MTT method.Furthermore,the resistance of Ophiopogon.japonicus polysaccharides to NDV was determined by different dosing methods.The results showed that the safe concentration of Ophiopogon japonicus polysaccharides on CEF was 78.125μg/mL,and the virus inhibition rate of OPS_(80) in the three different dosing methods was 37.36%,29.63%,39.30%,which were higher than that of the other 4 kinds.It's suggested that the Ophiopogon japonicus polysaccharides had obvious anti- NDV activity,among which the activity of OPS_(80) was the best,and the mechanism of action was most likely to be killing and blocking effect.
