PKCβ1和MEK1/2在子宫内膜异位症组织及间质细胞中的表达及意义

摘要 目的探讨蛋白激酶Cβ1(PKCβ1)和丝裂原激活的蛋白激酶激酶1/2(MEK1/2)在子宫内膜异位症(EMs)组织及间质细胞中的表达及其意义。方法收集因EMs行手术治疗于术中或术后经病理科证实为子宫内膜异位症的患者60例,分为EMs在位内膜组(30例)和EMs异位内膜组(30例);同期因良性卵巢肿瘤经手术治疗的患者30例作为对照组(正常子宫内膜组)。采用免疫组化法检测3组组织中PKCβ1,MEK1/2的表达;另选取EMs组在位内膜及正常组内膜各10例提取原代子宫内膜间质细胞分离培养,采用Western-blot,检测EMs组及正常组子宫内膜间质细胞中PKCβ1、MEK1/2、MMP9、PCNA蛋白的表达情况。结果EMs在位及异位内膜组中PKCβ1,MEK1/2的表达均高于正常子宫内膜组(P<0.05),且异位内膜组中PKCβ1,MEK1/2的表达高于在位内膜组(P<0.05)。EMs在位内膜组间质细胞中PKCβ1、MEK1/2蛋白的表达高于正常组间质细胞(P<0.05)。MMP9和PCNA是与细胞的侵袭和增殖性密切相关的蛋白,EMs在位内膜组间质细胞中MMP9、PCNA蛋白的表达高于正常组间质细胞(P<0.05),提示EMs间质细胞的侵袭和增殖性增高。相关分析发现PKCβ1、MEK1/2与MMP9、PCNA的表达水平呈正相关关系。结论PKCβ1、MEK1/2在子宫内膜异位症患者在位、异位内膜组织及在位内膜间质细胞中呈高表达,并与MMP9、PCNA的表达呈正相关,提示PKCβ1、MEK1/2可能在子宫内膜异位症间质细胞的侵袭、增殖中发挥作用。 Objective To investigate the expression and significance of protein kinase Cβ1(PKCβ1)and mitogen activated protein kinase 1(MEK1/2)in tissues and stromal cells of endometriosis.Methods 30 patients with endometriosis confirmed by pathology after operation.The patients with eutopic endometrium and eutopic endometrium were divided into EMs eutopic endometrium group and EMs ectopic endometrium group,and 30 patients with benign ovarian tumors were taken as control group(normal endometrium group).Immunohistochemical method was used to detect the expression of PKCβ1 and Magneto MEK1/2 in the tissues of the three groups.The primary endometrial stromal cells were isolated and cultured from 10 eutopic endometrium(EMs)and 10 normal endometrium.The expression of PKCβ1 and MEK1/2,MMP9,PCNA protein in endometrial stromal cells of EMs group and normal group were detected by western-blot.Results The expression of PKCβ1 and MEK1/2 in ectopic and eutopic endometrium was higher than that in normal endometrium(P<0.05),and the expression of PKCβ1 and MEK1/2 in ectopic endometrium was higher than those in eutopic endometrium(P<0.05).The expression of PKCβ1 and MEK1/2,MMP9,PCNA protein in eutopic endometrial stromal cells in EMs group was higher than that in normal stromal cells(P<0.05).Correlation analysis showed thatPKCβ1 and MEK1/2 was positively correlated with the expression level of PKCβ1 and MEK1/2 in eutopic endometrial stromal cells.Conclusion PKCβ1 and MEK1/2 are highly expressed in eutopic and ectopic endometrial tissues and eutopic endometrial stromal cells of patients with endometriosis,suggesting that PKCβ1 and MEK1/2 may play a role in the invasion and proliferation of endometriotic stromal cells.
