黄连解毒汤“异病同治”冠心病和2型糖尿病的网络药理学作用机制研究 被引量:11

摘要 目的:运用网络药理学方法,探究黄连解毒汤“异病同治”冠心病和2型糖尿病的共同作用机制。方法:通过中药系统药理学数据库平台(TCMSP)检索黄连解毒汤中4味药的所有化学成分及作用靶点,并通过Uniprot数据库进行基因的标准化处理;在OMIM和GeneCards数据库获取与冠心病和糖尿病相关的疾病靶点。利用Cytoscape3.7.2软件构建“药物-成分-靶点-疾病”网络图,运用STRING数据库构建蛋白互作PPI网络图,并运用Cytoscape3.7.2软件构建“药物-疾病”核心靶点网络图。利用DAVID在线数据库平台对黄连解毒汤治疗冠心病和2型糖尿病的共同靶点进行GO生物过程分析和KEGG通路富集分析。结果:检索得到黄连解毒汤活性成分103个,成分作用靶点140个,冠心病靶点5 342个,糖尿病靶点114个,药物与疾病的共同交集靶点14个,涉及AR、PPARG、TNF、IL6、CCL2、VEGFA、PON1等。GO生物过程分析共得到生物过程98个,细胞组分10个,分子功能10个,其中涉及positive regulation of gene expression(基因表达的正调控)、positive regulation of nitric oxide biosynthetic process(一氧化氮生物合成过程的正调控)、extracellular space(胞外空间)、cytokine activity(细胞因子活性)、steroid hormone receptor activity(类固醇激素受体活性)等生物过程;KEGG通路富集分析得到20条信号通路(P≤0.05),主要涉及到疟疾(Malaria)、癌症信号通路(pathways in cancer)、HIF-1信号通路(HIF-1 signaling pathway)、TNF信号通路(TNF signaling pathway)、NOD样受体信号通路(NOD-like receptor signaling pathway)、PI3K-Akt信号通路(PI3K-Akt signaling pathway)等。结论:黄连解毒汤“异病同治”冠心病和2型糖尿病具有多成分、多靶点、多通路的特点,为黄连解毒汤在临床中治疗冠心病和2型糖尿病提供理论依据,但黄连解毒汤治疗疾病的关键靶点和通路仍需进一步实验验证。 Objective:To explore the common mechanism of Huanglian Jiedu Decoction in treating coronary heart disease and type 2 diabetes by network pharmacology. Methods:All chemical components and targets of these four drugs in Huanglian Jiedu Decoction were retrieved through TCMSP,and the genes were standardized through Uniprot database. Disease targets related to coronary heart disease and diabetes were acquired from OMIM and Gene Cards databases. Network diagram of "drug-component-target-disease" was constructed by using the software of cytopscape 3.7.2,the PPI network diagram of protein interaction was constructed by using STRING database,and the network diagram of "drug-disease" core target was constructed by using the software of cytopscape 3.7.2. DAVID’s online database platform was used to analyze GO biological process and KEGG pathway enrichment of common targets of Huanglian Jiedu Decoction in treating coronary heart disease and type 2 diabetes. Results:A total of 103 active ingredients of Huanglian Jiedu Decoction were retrieved,including 140 acting targets,5 342 targets for coronary heart disease,114 targets for diabetes,and 14 intersection targets of drugs and diseases,involving AR,PPARG,TNF,IL6,CCL2,VEGFA,PON1,etc. The GO biological process analysis showed 98 biological processes,10 cell components and 10 molecular functions. It involved positive regulation of gene expression,positive regulation of nitric oxide biosynthesis process,extracellular space,cytokine activity,steroid hormone receptor activity and other biological processes. The enrichment analysis of KEGG pathway revealed 20 signal pathways(P≤0.05). It mainly involved malaria,pathway in cancer,HIF-1 signaling pathway,TNF signaling pathway,NOD-like receptor signaling pathway,PI3 K-Akt signaling pathway,etc. Conclusion:Huanglian Jiedu Decoction treats different diseases in the same way,and can be used to treat coronary heart disease and type 2 diabetes. It has the characteristics of multiple components,multiple targets and multiple pathways,which
