西黄丸对乳腺癌荷瘤小鼠肿瘤组织凋亡及相关因子表达的影响 被引量:5

Effect of Xihuang Pills(西黄丸)on Tumor Tissue Apoptosis and Related Factors Expression in Mice with Breast Cancer

摘要 目的:探讨西黄丸对乳腺癌细胞株MDA-MB-435移植瘤小鼠肿瘤生长及肿瘤组织凋亡的影响。方法:裸鼠皮下注射人乳腺癌细胞株MDA-MB-435建立肿瘤模型,35只荷瘤小鼠随机分为模型组、环磷酰胺组、西黄丸高剂量组、西黄丸中剂量组、西黄丸低剂量组,连续给药3周后脱颈椎处死,进行瘤体直径、瘤质量及脾脏指数的测定;采用HE染色、免疫组化染色及TUNEL染色方法观察肿瘤组织病理学变化、凋亡及相关蛋白表达的情况;采用RT-PCR方法检测肿瘤组织Bax mRNA、Bcl-2 mRNA及Caspase-3 mRNA表达。结果:与模型组比较,环磷酰胺组和西黄丸高剂量组小鼠肿瘤组织瘤质量显著降低(P<0.05);环磷酰胺组、西黄丸高剂量组和西黄丸中剂量组小鼠肿瘤组织凋亡区域明显增加(P<0.05);环磷酰胺组及西黄丸高剂量组小鼠肿瘤组织凋亡基因Bcl-2 mRNA和Bax mRNA的比率及Bcl-2 mRNA的表达显著下调(P<0.05),西黄丸低、中、高剂量组小鼠肿瘤组织凋亡基因Bax mRNA和Caspase-3 mRNA表达增加(P<0.05)。结论:西黄丸有抑制乳腺癌细胞株MDA-MB-435移植瘤裸鼠肿瘤生长的作用,可促进乳腺癌细胞株MDA-MB-435移植肿瘤组织凋亡,其机制可能是通过增加凋亡基因Bax mRNA和Caspase-3 mRNA的表达以促进肿瘤组织凋亡。 Objective:To investigate the effect of Xihuang Pills on tumor growth and tumor tissue apoptosis in mice transplanted with breast cancer cell line MDA-MB-435.Methods:Human breast cancer cell line MDAMB-435 was subcutaneously injected into nude mice to establish tumor model.Tumor-bearing mice were randomly divided into model group,cyclophosphamide group,Xihuang Pills high-dose group,Xihuang Pills medium-dose group and Xihuang Pills low-dose group.After 3 weeks of continuous administration,the cervical spine was sacrificed and the tumor diameter,tumor mass and spleen index were measured.HE staining,immunohistochemical staining and TUNEL staining were used to observe the pathological changes,apoptosis and related protein expression of tumor tissues.The expressions of Bax mRNA,Bcl-2 mRNA and Caspase-3 mRNA in tumor tissues were detected by RT-PCR.Results:Compared with the model group,the tumor mass of mice in the cyclophosphamide group and the Xihuang Pills high-dose group was significantly reduced(P<0.05).The apoptotic area of tumor tissue in cyclophosphamide group,Xihuang Pills high-dose group and the Xihuang Pills mediumdose group increased significantly(P<0.05).The ratio of apoptosis genes Bcl-2 mRNA and Bax mRNA and the expression of Bcl-2 mRNA in the tumor tissues of mice in the cyclophosphamide group and the Xihuang Pills high-dose group were significantly down-regulated(P<0.05).The expression of apoptosis genes Bax mRNA and Caspase-3 m RNA in tumor tissues of mice in Xihuang Pills low,medium and high-dose group increased(P<0.05).Conclusion:Xihuang Pills can inhibit the tumor growth of breast cancer cell line MDA-MB-435 transplanted tumor in nude mice,and can promote the apoptosis of breast cancer cell line MDA-MB-435 transplanted tumor tissue.The mechanism may be to promote the apoptosis of tumor tissue by increasing the expression of Bax m RNA and caspase-3 m RNA.
