摘要
建立了在碘化钾-淀粉体系中利用分光光度计测定肉制品中微量亚硝酸钠的方法。试验基于在酸性氯化钠介质中,碘化钾与亚硝酸钠反应析出的碘能与淀粉形成蓝色配合物的原理,优化反应体系中的各项参数。结果表明,在测定波长为581nm、2.00mL盐酸溶液(1.0mol/L)与5.00mL氯化钠溶液(200g/L)形成的溶液环境中,10.00mL样品溶液与1.00mL碘化钾溶液(5g/L)、1.00mL淀粉溶液(1%)以及1.00mL磷酸缓冲溶液(pH6.86),振摇反应20min,生成的蓝色配合物有最大吸光度。亚硝酸钠溶液在0.05~0.50μg/mL浓度范围内线性良好,相关系数R2为0.999,检出限为0.002mg/L。加标试验分别添加4、20和100μg/L3个水平,加标回收率达到84.9%~94.6%。该方法该高效、灵敏、准确,可用于肉制品中微量亚硝酸钠的测定。
A method for the determination of sodium nitrite in meat products by potassium iodide-starch system spectrophotometry was established.The principle of this experiment is that when potassium iodide reacts with sodium nitrite in the medium of acid sodium chloride,iodine will be precipitated,and adding starch into the system,a blue complex will be observed.The results showed that 10.00 mL sample solution was mixed with 2.00mL HCl(1.0mol/L)and 5.00mL NaCl(200g/L)and detection was carried out at wavelength 581 nm.After adding 1.00 mL KI(5g/L),1.00mL starch(1%)and 1.00 mL phosphoric acid buffer(pH6.86)and shaking for 20 minute,the blue complex reached its maximum absorbance.The linear relationship of sodium nitrite was in the concentration range of 0.05~0.50μg/mL.The correlation coefficient R2 was 0.999 and the detection limit was 0.002 mg/L.The standard tests were performed at three levels of 4,20 and 100μg/L,and the recovery rate was 84.9%~94.6%.This method was highly effective,sensitive and accurate.It can be used to determine the content of sodium nitrite in meat products.
