酸浆宿萼中苦瓜甙的提取分离纯化及应用研究 被引量:1

摘要 通过溶剂法对亚临界法提取酸浆宿萼中类胡萝卜素实验的残渣进行提取实验得到苦瓜甙,并进行分离纯化和功能性产品的开发。提取所采用的实验条件为:乙醇浓度70%,提取温度50℃,提取时间3h,提取次数三次,料液比1∶15。结果表明苦瓜甙浸膏出品率为18.74%,苦瓜甙的含量为6.23%,苦瓜甙得率为1.17%。提取得到的苦瓜甙先后经过醇沉法、旋蒸、重新溶解、柱层析等过程进行纯化。实验过程中分别选择乙醇浓度为85%、87.5%、90%、92.5%、95%来进行实验,通过对比得出纯化效果最好的乙醇浓度。对实验结果进行综合考虑,选定乙醇浓度90%为苦瓜甙纯化过程的最佳乙醇浓度,此时洗脱剂为70%乙醇,洗脱速度为5mL/min。在此条件下,纯化后的苦瓜甙含量为78.10%,比起纯化前的10%有非常明显的提升,此时苦瓜甙的得率为84.90%,符合工业生产的要求。实现了具有降血糖功效的苦瓜甙片剂的开发。 Charantin was extracted from residues after carotenoids extracted from calyxes of physalis by subcritical fluid extraction technology.The experimental conditions are as follows:ethanol concentration 70%,extraction temperature 50℃,extraction 3h for 3 times,and ratio of material-liquid 1∶15.The results show that the yield of charantin extract is 18.74%,the content of charantin is 6.23%,and the yield of charantin is 1.17%.Then the extracts are purified by alcohol precipitation,evaporation,redissolution and column chromatography.Ethanol concentration was selected at 85%,87.5%,90%,92.5%,95%,the optimum ethanol concentration is 90%based on purification effects,the eluent is 70%ethanol and elution rate is 5mL/min.Under the above conditions,the content of charantin is 78.10%,a significant increase from 10%before purification.The yield of charantin is 84.90%,which meets the requirements of industrial production.A hypoglycemic charantin tablet is developed.
