吡非尼酮对糖尿病肾病小鼠肾纤维化的改善作用及机制研究 被引量:1

摘要 目的探究吡非尼酮对糖尿病肾病小鼠肾纤维化的改善作用及相关机制。方法将30只雄性小鼠随机分为对照组、模型组、实验组,各10只。模型组与实验组小鼠采用腹腔注射链尿佐菌素的方法建立糖尿病肾病模型,造模成功后,对照组及模型组小鼠灌胃1%羧甲基纤维素钠溶液,实验组小鼠给予吡非尼酮溶液灌胃,均连续灌胃7 d。比较三组小鼠肾功能水平,肾组织Masson染色情况及肾组织TGF-β、α-SMA、ColⅠ蛋白相对表达水平。结果与对照组比,模型组小鼠24 h尿蛋白含量、BUN、SCr水平及肾组织TGF-β、α-SMA、ColⅠ蛋白相对表达水平均升高,且实验组显著低于模型组(P<0.05);HE染色结果显示,对照组小鼠肾小管间质无明显纤维化,未见增生及炎性细胞浸润;模型组小鼠肾小管间质有明显增生、纤维化,伴有炎性细胞浸润;实验组小鼠肾小管间质有轻微纤维化,且有少量炎性细胞浸润。结论吡非尼酮可改善糖尿病肾病小鼠肾功能及肾纤维化,同时减少小鼠肾组织TGF-β、α-SMA、ColⅠ蛋白表达,从而控制肾纤维化的发展进程。 Objective To investigate the effect of pirfenidone on renal fibrosis in diabetic nephropathy mice and its related mechanism.Methods 30 male mice were randomly divided into the control group,the model group and the experimental group,each group with10 mice.The diabetic nephropathy model was established by intraperitoneal injection of streptozotocin.After successful modeling,the mice in the control group and the model group were given 1%sodium carboxymethy cellulose solution by gavage,the mice in the experimental group were given pirfenidone solution by gavage,and all mice were given gavage for 7 day.The level of renal function and Masson staining of renal tissue,and relative expression levels of TGF-β,α-SMA and ColⅠof renal tissue were compared among the three groups.Results Compared with the control group,the 24 h urinary protein content,BUN,SCr levels and the relative expression levels of TGF-β,α-SMA and ColⅠprotein in the model group and the experimental group were increased,and the experimental group was significantly lower than the model group(all P<0.05).The results of HE staining showed that,there was no obvious fibrosis in the renal tubulointerstitium of the control group,no proliferation and inflammatory cell infiltration were found.In the model group,renal tubulointerstitial hyperplasia,fibrosis and inflammatory cell infiltration were observed.In the experimental group,there was slight fibrosis and a small amount of inflammatory cells infiltration.Conclusion Pirfenidone can improve renal function and renal fibrosis in diabetic nephropathy mice,at the same time,the quantity and expression of TGF-β,α-SMA and ColⅠof renal tissue were decreased,so as to control the development process of renal fibrosis,improve the therapeutic effect.
