河北省肾综合征出血热疫区鼠携带汉坦病毒基因特征分析 被引量:5

Genetic characteristics of Hantavirus carried by rats in the main areas where hemorrhagic fever with renal syndrome is endemic in Hebei Province,China

摘要 目的了解河北省肾综合征出血热(HFRS)疫区鼠携带汉坦病毒(HV)的基因特征。方法收集2019年河北省各地市捕获的鼠类标本,无菌下解剖取肺组织,应用Real-time PCR对阳性鼠标本进行HV G2片段扩增并测序,运用DNASTAR 7.1软件包对核苷酸序列进行同源性比较,采用MEGA 5.22软件构建系统发生树分析。结果共收集951份鼠肺标本,经Real-time PCR检测HV阳性标本共24份,均为汉城型(SEO)。对12份标本进行序列测定,鼠肺标本病毒间变异不大,同源性达97.3%~100.0%。新测序株与以往测序株比较同源性为97.3%~99.7%。系统发生树分析显示12份标本均为S3亚型HV。结论河北省HFRS疫区HV基因型以SEO型为主,亚型为S3亚型,同亚型病毒基因高度同源,无明显变异,遗传物质具有区域稳定性特征。 Objective To ascertain the genetic characteristics of Hantavirus(HV)carried by rats in the endemic foci for hemorrhagic fever with renal syndrome(HFRS)in Hebei Province.Methods Rats were collected in Hebei Province in 2019.Lung tissues were isolated in a sterile environment.Fragments of the G2region of HV were amplified using realtime PCR and sequenced.The software DNASTAR 7.1was used to compare nucleotide homology and MEGA 5.22was used to construct phylogenetic trees.Results A total of 951samples were collected,and 24HV-positive specimens were detected with real-time PCR,all of which were SEO types.Twelve lung samples were selected for sequencing.HV displayed slight variation,and its nucleotide sequences were 97.3%-100%identical.It was 97.3%-99.7%identical to other strains previously isolated in Hebei Province.Phylogenetic analysis indicated that all of the strains are grouped into the same S3subtype of SEO.Conclusion SEO was the major epidemic genotype of HV in Hebei Province,and S3was the major subtype.HV belonging to the same subtype was highly identical and varied slightly.Its genetic material is regionally stable.
