摘要
目的:研究聚腺苷二磷酸核糖聚合酶(PARP)抑制剂鲁卡帕尼对于鱼藤酮诱导的SH-SY5Y细胞凋亡的影响。方法:将呈对数生长的SH-SY5Y细胞分为3组:对照组(正常培养)、模型组(鱼藤酮10μmol/L诱导)、药物组(鲁卡帕尼5μmol/L预处理1小时后加入鱼藤酮10μmol/L),用CCK8检测各组细胞的存活率,用蛋白质印迹(western blot)技术检测各组凋亡相关蛋白bcl-2和bax的蛋白表达量,用流式细胞仪检测各组细胞的凋亡率。结果:与对照组相比,模型组的细胞存活率明显下降,bcl-2蛋白表达量降低,细胞凋亡率上升,促凋亡蛋白bax表达量增多。与模型组相比,鲁卡帕尼预处理组细胞存活率上升,bcl-2蛋白表达量增多,细胞凋亡率下降,促凋亡蛋白bax表达量减少。差异有统计学意义(P<0.05)。结论:PARP抑制剂鲁卡帕尼可以抑制鱼藤酮诱导的SH-SY5Y细胞凋亡。
Objective:To investigate the effect of poly-ADP-ribose polymerase(PARP)inhibitor rucaparib on apoptosis of SH-SY5 Y cells induced by rotenone.Methods:SH-SY5 Y cells were cultured and divided into 3 groups,normal cultured SH-SY5 Y cells were used as control group,model group was treated with DMEM containing 10μmol/L rotenone,drug group was treated with DMEM containing 10μmol/L rotenone after 5μmol/L rucaparib 1 hour.The survival rate,apoptosis rate of SH-SY5 Y cells,and expression of bcl-2 and bax protein in different groups were compared.Results:The survival rate of SH-SY5 Y cells and the expression level of bcl-2 were lower in model group than that in control group,while the expression level of bax and the apoptosis rate were higher in model group than in control group.The survival rate of SH-SY5 Y cells and the expression level of bcl-2 were higher in rucaparib group than that in model group,while the expression level of bax and the apoptosis rate were lower in rucaparib group than in model group.Conclusion:poly-ADPribose polymerase(PARP)inhibitor rucaparib can relief apoptosis of SH-SY5 Y cells induced by rotenone.
