摘要
生长分化因子8(Growth differentiation factor 8,GDF-8)在肌肉的分化和生长中有负调控作用,但其是否参与调节动物生殖活动证据仍然不足。为研究GDF-8蛋白在牦牛发情周期输卵管不同部位中的表达差异,从屠宰场采集卵泡期和黄体期牦牛输卵管峡部、伞部和壶腹部组织样品,进行了免疫组织化学染色和Western Blotting检测。结果表明:在牦牛卵泡期和黄体期输卵管的肌层细胞和黏膜上皮细胞中均有GDF-8阳性细胞分布,特别是壶腹部和伞部黏膜上皮细胞表达GDF-8蛋白较丰富;Western Blotting结果表明,GDF-8蛋白在牦牛卵泡期和黄体期输卵管的峡部、壶腹部和伞部也均有表达,但存在表达差异,卵泡期壶腹部和黄体期输卵管伞部表达量明显高于其他部位。综上所述,GDF-8蛋白在输卵管黏膜上皮细胞表达较为丰富,提示GDF-8在哺乳动物生殖过程中发挥重要作用。
Growth differentiation factor 8(GDF-8)plays a negative role in the regulation of muscle differentiation and growth,but whether it involves in reproduction is not clear yet.The purpose of this study was to investigate the expression of GDF-8 protein in different parts of oviduct during estrous cycle of yak.The samples of isthmus,fimbria and ampulla of oviduct in follicular and luteal phase were collected from slaughterhouse.Immunohistochemical staining showed that GDF-8 positive cells were distributed in myometrial layer and mucosal layer of oviduct in follicular and luteal phases,especially in ampulla and fimbria.The expression of GDF-8 protein was abundant in the mucosal epithelial cells.Western Blotting results showed that GDF-8 protein was also expressed in isthmus,ampulla and fimbria of oviduct in follicular and luteal phases of yak,but there was difference in expression.The expression of GDF-8 protein in ampulla and fimbria of oviduct in follicular phase were significantly higher than that in other parts of oviduct.In conclusion,GDF-8 protein was highly expressed in oviduct mucosal epithelial cells,which points out that GDF-8 could play an important role in mammalian reproduction.
