铁皮石斛多糖对H_(2)O_(2)损伤HaCaT细胞的保护作用 被引量:5

The Protection Effect of Dendrobium Officinale Polysaccharides on H_(2)O_(2)-induced Oxidative Stress in HaCaT Cells

摘要 目的考察铁皮石斛多糖对表皮角质细胞氧化损伤的保护作用及机理,为铁皮石斛多糖的有效应用提供参考。方法筛选出不影响HaCaT细胞正常生长的铁皮石斛多糖浓度,以及有效损伤HaCaT细胞的H_(2)O_(2)浓度,观察H_(2)O_(2)对HaCaT细胞存活率和抗氧化相关酶的影响。结果 500μmol·L^(-1)的H_(2)O_(2)孵育HaCaT细胞2 h,细胞相对存活率为67.25%,50μg·mL^(-1)的铁皮石斛多糖能让HaCaT细胞的相对存活率恢复至90.14%;铁皮石斛多糖可以通过提高HaCaT细胞SOD、CAT活性及GSH含量,降低MDA含量,从而减轻H_(2)O_(2)对HaCaT细胞的氧化损伤作用。结论铁皮石斛多糖对H_(2)O_(2)损伤HaCaT细胞的保护作用可能通过提高HaCaT细胞相关抗氧化酶的活性、降低细胞脂质过氧化程度来实现。 OBJECTIVE To explore the protection effect and functional mechanisms of polysaccharides of Dendrobium Officinale polysaccharides on oxidative stress to human epidermal keratinocytes, in order to provide the scientific evidence of its effective application. METHODS The concentration of polysaccharides from Dendrobium Officinale without affecting the normal growth of HaCaT cells and the concentration of H_(2)O_(2) induced oxidative damage in HaCaT cells were screened out to investigate the relative survival rate of HaCaT cells and the activity of related anti-oxidative enzymes. RESULTS The relative survival rate of HaCaT cells treated with 500 μmol·L^(-1) H_(2)O_(2) for 2 h was 67.25%,while Dendrobium Officinale polysaccharides exhibited distinct protection effect on H_(2)O_(2)-induced oxidative stress in HaCaT cells.The relative survival rate of HaCaT cells treated with 50 μg·mL^(-1) polysaccharides before treated with 500 μmol·L^(-1) H_(2)O_(2) could be restore to 90.14%.The activities of superoxide dismutase(SOD),catalase(CAT) and the contents of glutathione(GSH) increased while malondialdehyde(MDA) decreased in HaCaT cells after treated with polysaccharides significantly, compared to the cells only treated with H_(2)O_(2).Thus, the oxidative damage of H_(2)O_(2) to HaCaT cells were alleviated. CONCLUSION The protection effect of Dendrobium Officinale polysaccharides on H_(2)O_(2)-induced oxidative stress may be related to promoting the activity of antioxidative enzymes and reducing the degree of lipid peroxidation of HaCaT cells.
