摘要
为了解彭泽鲫(Carassius auratus var.pengsenensis)♀×兴国红鲤(Cyprinus carpio var.singuonensis)♂杂交四倍体子代的细胞遗传特性,本研究以彭泽鲫♀×兴国红鲤♂杂交四倍体子代为研究对象,以彭泽鲫雌核发育子代为内参,采用流式细胞仪测量其DNA相对含量,并对彭泽鲫雌核发育子代与杂交四倍体子代进行红细胞形态、大小的比较,另取杂交四倍体子代肌肉、鳍条组织块进行细胞培养,制作染色体标本,对其进行染色体计数及核型分析。结果显示:杂交四倍体子代仍为四倍体,其红细胞长轴、表面积、体积、核长轴、核表面积、核体积均显著大于彭泽鲫雌核发育子代。从形态上看,杂交四倍体子代红细胞相较于彭泽鲫雌核发育子代更长,辅助证明杂交四倍体子代为四倍体。杂交四倍体子代染色体组由200+条染色体组成,核型公式为4n=42m+64sm+62st+32t,NF=368,多出的染色体为超数染色体,未发现有异型性染色体,杂交四倍体子代属于中位演化类群。综上表明,彭泽鲫♀×兴国红鲤♂杂交四倍体子代仍为四倍体。
This study used Carassius auratus var.pengsenensis♀×Cyprinus carpio var.singuonensis♂hybrid tetraploid progeny as the research object and C.auratus var.pengsenensis gynogenetic progeny as internal controlto understand the cytogenetic characteristics of C.auratus var.pengsenensis♀×C.carpio var.singuonensis♂hybrid tetraploid progeny and explore the development and application prospects of new varieties.Meanwhile,flow cytometry was used to measure the relative DNA content.For C.auratus var.pengsenensis gynogenetic progeny and hybrid tetraploid progeny,the red blood cell morphology and size were compared in the generations.The muscle and fin ray tissue blocks of the hybrid tetraploid progeny were taken for cell culture,chromosome specimens were made,and chromosome count and karyotype analysis were performed on them.The results showed that the progeny of hybridized tetraploid was still tetraploid.The long axis,surface area,volume,long axis,surface area and volume of red blood cells of hybridized tetraploid progeny were significantly larger than those of C.auratus var.pengsenensis′s female nuclear progeny.From the morphological point of view,the red blood cells of hybridized tetraploid progeny were longer than those of C.auratus var.pengsenensis′s female nuclear progeny,which proved that hybridized tetraploid progeny was tetraploid.The hybridized tetraploid progeny was tetraploid.The karyotype formula was 4N=42m+64sm+62st+32t,NF=368.The extra chromosomes weresupernumerary chromosomes,no atypical sex chromosomes were found.The offspring of hybrid tetraploid belonged to the median evolution group.This study identified C.auratus var.pengsenensis♀×C.carpio var.singuonensis♂hybrid tetraploid progeny as tetraploid,which provideda theoretical basis for subsequent germplasm identification and hybrid breeding,and also provided data support for in situ hybridization and gene mapping.
