全自动微波辅助快速组织处理对免疫组化染色的影响 被引量:5

摘要 目的探讨对不同脏器组织使用全自动微波辅助快速组织处理,对其免疫组化染色的影响分析。方法收集解放军总医院病理科自2018-03—2018-05,临床送检的不同脏器的常规手术切除标本130例,充分固定后,在每例标本相同部位取材,大小1.5 cm×1.5 cm×0.2 cm的组织2块,同时分成2组,第一组使用常规全自动脱水机处理作为对照组;另外一组使用全自动微波辅助快速组织处理仪进行快速组织处理,作为实验组。应用免疫组织化学(IHC)方法,对两组组织块,按不同类型组织选择了相应的8种抗体(Ki-67、CK、CD31、SMA、ER、PR、CD3、CD20)进行染色,通过盲法,镜下观察,评价切片染色质量,对比其差异。结果 130例实验组与对照组的组织,进行免疫组化染色后,两组切片的合格率分别为96.9%和95.0%,χ^(2)=1.241,P=0.265(P>0.05);所选择的8种抗体染色,阳性定位准确,着色、分布均匀,背景清晰,阳性表达基本一致。两种方式处理后的组织,免疫组化染色结果无显著差异。结论对固定充分的不同脏器标本,使用全自动微波辅助快速组织处理仪进行快速组织处理,可高效缩短组织处理时间,对免疫组化染色结果无明显影响,可用于免疫组化鉴别诊断。 Objective To investigate the effect of the automatic microwave-assisted rapid tissue processing on the immunohistochemical staining in specimens of different viscera. Methods A total of 130 normal tissue samples of different viscera were collected from the PLA General Hospital during March 2018 to May 2018. After fixation,2 objective tissues were obtained from the same part of the viscera and divided into 2 groups. Size of the tissues were below 1. 5 cm ×1. 5 cm × 0. 2 cm. Group 1 was treated as control and the traditional tissue processor was used to be hydro-extracted,while Group 2 was treated with automatic microwave-assisted rapid tissue processor for rapid processing. Two tissues were stained according to several antibodies as Ki-67,CK,CD31,ER,PR,CD3,CD20 and SMA. Then,the quality of the tissue staining was assessed via blind test by microscopy. Results The 130 tissue samples of control and test groups were stained and the qualified rate was 96. 9% and 95. 0% respectively( χ^(2)= 1. 241,P>0. 05). Localization of 8 antibodies in the tissue samples were accurate,staining even and the background was clear. Meanwhile,it had good accordance between two positive groups. There was no significant difference in immunohistochemical staining results between the two methods.Conclusion Automatic microwave-assisted rapid tissue processor to treat the fixed tissues can not only effectively shorten the processing time,but also have no obvious effect on immunohistochemical staining results compaired with the traditional method;the sections are qualified and suited for diagnosis.
