摘要
喜树因能够合成具有抗癌作用的喜树碱及其衍生物而备受关注。喜树叶片是喜树碱积累的主要场所,且在表皮毛中大量积累。因此,深入了解表皮毛发育调控对于喜树碱合成调控具有重要意义。然而,目前对于喜树叶片表皮毛研究报道较少。本研究基于模式植物拟南芥表皮毛发育的已有研究,分析了外源激素6-BA对喜树叶片表皮毛相关基因转录水平的影响。用6-BA (2.2μmol/L)处理喜树叶片后,喜树表皮毛发育相关基因中CaGIS、CaTTG1、CaMYB23、CaGL2、CaEGL3、CaSPL9、CaTCL1的转录水平被上调,尤其是CaGL2和CaEGL3,其实验组分别为对照组的5倍和4.5倍。再将此结合表皮毛发育模型推测,6-BA能够提高喜树正调控因子CaGIS的转录水平,进而调控转录复合体CaMYB23-CaGL3-CaTTG1,随后激活喜树表皮毛发育下游基因CaGL2,最终激活表皮毛的启动。本研究探讨了6-BA对喜树表皮毛相关基因转录水平的调控,对于研究喜树表皮毛的发育和提高喜树碱积累具有重要的应用价值。
Camptothecin has attracted much attention because it can synthesize camptothecin and its derivatives with anti-cancer effects.Camptotheca acuminata leaves is the main site for the accumulation of camptothecin that massively accumulates in the epidermis.Therefore,understanding the regulation of trichome development is of great significance for the regulation of camptothecin biosynthesis.However,there are few reports on the trichome of Camptotheca acuminata leaves.In this study,we have analyzed the effects of the exogenous hormone 6-BA on the transcription level of trichome-related genes in leaves of Camptotheca acuminata based on existing studies on the development of trichome in the model plant Arabidopsis.After leaves of Camptotheca acuminata were treated with 6-BA(2.2μmol/L),the transcriptions of CaGIS,CaTTG1,CaMYB23,CaGL2,CaEGL3,CaSPL9,and CaTCL1 were increased in trichome development related genes of Camptotheca acuminata,especially CaGL2 and CaEGL3 whose levels of experimental group were 5 and 4.5 times as high as those of the control group.Coupled with trichome development model,we conclude that 6-BA can improve the transcription of Camptotheca acuminata’s positive regulator CaGIS to regulate the CaMYB23-CaGL3-CaTTG1 complex,then activates CaGL2,a gene downstream of tri chome development in Camptotheca acuminata,and eventually activates the initiation of trichome.Regulation of 6-BA on the transcription level of trichome development-related genes in Camptotheca acuminata leaves is studied here,which has important application value for studying the development of trichome of Camptotheca acuminata and improving the accumulation of camptothecin.
