摘要
目的:探讨用实时荧光定量多聚酶链反应(Real-time PCR)扩增技术联合浓缩涂片染色法快速诊断结核病的效果。方法:选择高州市人民医院检验科收到的526份分泌物标本作为研究对象。对这526份分泌物标本分别采用直接涂片染色法、Real-time PCR扩增技术联合浓缩涂片染色法进行病原体检测。然后,观察采用BACTEC MGIT-960培养法、直接涂片染色法、Real-time PCR扩增技术联合浓缩涂片染色法对526份分泌物标本进行病原体检测的时间、结核分枝杆菌的检出率及用Real-time PCR扩增技术联合浓缩涂片染色法诊断结核病的灵敏度、特异度及准确率。结果:与采用直接涂片染色法相比,采用BACTEC MGIT-960培养法、Real-time PCR扩增技术联合浓缩涂片染色法对这526份分泌物标本进行病原体检测时结核分枝杆菌的检出率更高,P<0.05。与采用BACTEC MGIT-960培养法相比,采用直接涂片染色法、Real-time PCR扩增技术联合浓缩涂片染色法对这526份分泌物标本进行病原体检测的时间更短,P<0.05。采用Real-time PCR扩增技术联合浓缩涂片染色法诊断结核病的灵敏度、特异度、准确率分别为89.7%、94.3%、93.0%。结论:用Real-time PCR扩增技术联合浓缩涂片染色法可快速对结核病患者的病情进行诊断,且准确率较高。
Objective:To investigate the effect of real-time quantitative PCR amplification combined with concentrated smear staining in the rapid diagnosis of tuberculosis(TB).Methods:526 secretion specimens from the laboratory of Gaozhou People’s Hospital were selected as the research object.The 526 secretions were detected by direct smear staining and Real-time PCR amplification combined with concentrated smear staining,respectively.And then,The time of pathogen detection and the detection rate of Mycobacterium tuberculosis in 526 secretion specimens were observed by Bactec MGIT-960 culture method,direct smear staining method,Real-time PCR amplification combined with concentrated smear staining method Sensitivity,specificity and accuracy of PCR amplification combined with concentrated smear staining in the diagnosis of tuberculosis.RESULTS:Compared with the direct smear staining method,the detection rate of Mycobacterium tuberculosis in the 526 secretion specimens was higher when Bactec MGIT 960 culture method,Real-time PCR amplification technology combined with concentrated smear staining method,P<0.05.Compared with BACTEC MGIT-960 culture method,the time of direct smear staining,real-time PCR amplification combined with concentrated smear staining for pathogen detection in 526 secretion specimens was shorter(P<0.05).The sensitivity,specificity and accuracy of real-time PCR amplification combined with concentrated smear staining were 89.7%,94.3%and 93.0%,respectively.Conclusion:Real-time PCR amplification combined with concentrated smear staining can quickly diagnose the disease of tuberculosis patients with high accuracy.
