摘要 代谢相关脂肪性肝病(MAFLD)正在成为全球最主要的慢性肝病病因,全球约25%的人口患有不同程度的MAFLD。MAFLD发病机制复杂,美国食品药品监督管理局(FDA)及欧盟药品管理局(EMA)目前尚未批准任何药物用于MAFLD的治疗。Kupffer细胞是肝脏特有的巨噬细胞,占人体定殖巨噬细胞的80%~90%,可通过旁分泌作用调控肝细胞的生理功能,在维持肝脏稳态中发挥重要作用。Kupffer细胞是具有免疫原性的巨噬细胞,在不同微环境的影响下可以分化为M1和M2型,其参与MAFLD发病机制中脂肪变性、氧化应激及纤维化等多个进程。本文综述了Kupffer细胞在MAFLD发病机制中的多种作用,旨在为针对Kupffer细胞设计MAFLD的多靶点协同治疗策略提供相关思路。 Metabolic associated fatty liver disease(MAFLD)is emerging as the leading cause of chronic liver disease in the world,with about 25%of the global population suffering from different degree of MAFLD.The pathogenesis of MAFILD is complex,and the United States food and drug administration(FDA)and the European medicines agency(EMA)have not yet approved any drug for the treatment of MAFILD.Kupffer cells are liver specific macrophages,accounting for 80%~90%of human colonized macrophages,which can regulate the physiological function of liver cells through parocrine action and play an important role in maintaining liver homeostasis.Kupffer cells are immunogenic macrophages,which can differentiate into M1 and M2 types under the influence of different microenvironments.Kupffer cells are involved in the pathogenesis of MAFLD,such as steatosis,oxidative stress and fibrosis.This paper reviews the various roles of Kupffer cells in the pathogenesis of MAFLD,aiming to provide some ideas for designing the multi-target cooperative therapy strategy for Kupffer cells.
