摘要
目的:以无水乙醇建立小鼠急性胃溃疡出血模型,评价不同剂型艾速平对胃溃疡的保护作用。方法:将30只雌性昆明小鼠随机分为对照组、胃溃疡模型组、高、低剂量艾速平注射液组(6.06 mg·kg^(-1)、3.03 mg·kg^(-1))、高、低剂量艾速平肠溶胶囊组(6.06 mg·kg^(-1)、3.03 mg·kg^(-1))(n=5)。除对照组外,所有小鼠预防性给药5 d后灌胃无水乙醇0.2 mL建立急性胃溃疡模型。1 h后处死小鼠,观察胃黏膜形态并计算溃疡指数;同时HE染色胃粘膜损伤;收集胃液以pH试纸测定小鼠胃液pH值,采用无机磷测定法测定胃蛋白酶活性。结果:无水乙醇明显诱导小鼠胃粘膜溃疡、出血,溃疡指数与对照组相比明显升高(P<0.05),且胃蛋白酶活性显著升高(P<0.05),然而未见胃液pH值改变(P>0.05);低剂量艾速平肠溶胶囊、高剂量艾速平注射液、低剂量艾速平注射液治疗均可明显减少无水乙醇诱导的胃粘膜溃疡、出血,且胃蛋白酶活性显著降低(P<0.05),其中低剂量艾速平注射液治疗后溃疡指数与模型组相比明显降低(P<0.05);其余剂型及剂量组未见明显改变(P>0.05)。结论:口服艾速平肠溶胶囊3.03 mg·kg^(-1)、腹腔注射艾速平注射液(6.06 mg·kg^(-1)、3.03 mg·kg^(-1))对乙醇致小鼠急性胃溃疡出血模型具有一定的保护作用,可减轻溃疡形成、抑制胃蛋白酶活性。
Objective:Mice model of acute gastric ulcer was established with anhydrous ethanol to evaluate the protective effects of different dosage forms of esomeprazole sodium on gastric ulcer.Methods:Thirty female Kunming mice were randomly assigned into six groups including control group,gastric ulcer model group,high and low dose of esomeprazole sodium injection group(6.06 mg·kg^(-1),3.03 mg·kg^(-1))and high and low dose of esomeprazole sodium enteric capsules group(6.06 mg·kg^(-1),3.03 mg·kg^(-1))(n=5).Except the control group,all mice were intragastrically administered with anhydrous ethanol 0.2 mL to establish the acute gastric ulcer model after saline(model group)or esomeprazole sodium(all treatment groups)preventive administration for 5 days.One hour later,all mice were sacrificed to observe gross morphology of gastric mucosa.The pathological changes of gastric tissue were observed under microscope,and gastric juice pH and pepsin activity were measured.Results:Compared with the control group,anhydrous ethanol significantly induced gastric mucosal ulcer and bleeding in mice(P<0.05),and the pepsin activity was significantly increased(P<0.05),however,no change in gastric juice pH was observed(P>0.05).The gastric mucosal ulcers and bleeding induced by anhydrous alcohol were significantly reduced after treatment with esomeprazole sodium enteric capsules and esomeprazole sodium for injection,and the pepsin activity was significantly decreased(P<0.05),and the pathological scores were significantly decreased after treatment with esomeprazole sodium for injection compared with the model group(P<0.05).However,other dosage forms and dose groups showed no significant changes(P>0.05).Conclusions:Oral administration of esomeprazole sodium enteric capsules at 3.03 mg·kg^(-1) and intraperitoneal injection of esomeprazole sodium had a certain protective effect on the mice model of acute gastric ulcer bleeding induced by ethanol,which could reduce the formation of ulcer and inhibit the activity of pepsin.
