梵净山地区中华蜜蜂种群遗传分析 被引量:3

摘要 由于本土中华蜜蜂种群面临多种威胁,有必要了解自然保护区在蜜蜂遗传资源保护方面的作用。利用15个微卫星分子标记,分析了贵州梵净山国家级自然保护区(保护区)及周边区域的中华蜜蜂遗传结构。结果表明,在遗传分化上,保护区内外的中华蜜蜂没有产生种群遗传分化;在遗传多样性上,保护区内中华蜜蜂的等位基因数(N_a)为3~20、期望杂合度(H_(e))为0.689 1±0.040 5、香农指数(I)为1.508 6±0.133 3、多态信息含量(PIC)为0.647 3±0.170 9。保护区外中华蜜蜂的N_(a)为3~20,H_(e)为0.691 4±0.037 1、I为1.512 4±0.127 7、PIC为0.647 4±0.163 4。保护区内外的中华蜜蜂维持了高水平的遗传多样性,估测了其有效种群大小为8 171.2。保护区及周边区域在本土蜜蜂遗传资源保护上发挥了积极作用,使得中华蜜蜂能够维持较大的种群数量和较高的遗传多样性。 Nature reserves play a crucial role in wildlife and ecosystem conservation.A local honeybee,Apis cerana cerana,is threatened by several factors,and it is necessary to understand the role of nature reserves in the conservation of genetic resources of Apis cerana cerana.In this study,population genetics analysis was conducted for honeybees from the Guizhou Fanjingshan National Nature Reserve and its surroundings through 15 microsatellite markers.The results revealed that there was no population genetic difference between the honeybees inside and outside of Fanjingshan.As for genetic diversity,the number of alleles(N_(a))ranged from 3 to 20,the expected heterozygosity(H_(e))was 0.6891±0.0405,Shannon index(I)was 1.5086±0.1333,and polymorphic information content(PIC)was 0.6473±0.1709 for honeybees inside Fanjingshan,while N_(a) was 3~20,He was 0.6914±0.0371,I was 1.5124±0.1277,and PIC was 0.6474±0.1634 for honeybees outside Fanjingshan.Honeybees from both inside and outside Fanjingshan showed high level of genetic diversity,and the effective population size was approximately 8171.2.We speculate that Fanjingshan Nature Reserve and its surrounding have played a positive role in the conservation of local honeybees,facilitating the maintenance of a large honeybee population with high genetic diversity.
