摘要
目的:探讨原发性肝癌组织中树突状细胞特异性细胞间黏附分子-3结合非整合素(DC-SIGN)表达与临床价值。方法:收集2018年3月至2019年6月贵州省人民医院的40例接受手术切除原发性肝癌患者,采集手术切除肝癌组织样本、癌旁肝组织样本各40份。分别通过免疫组织化学法检测样本中DC-SIGN蛋白表达以及通过反转录-聚合酶链反应(RT-PCR)方法检测样本DC-SIGN mRNA表达;分析肝癌组织样本中DC-SIGN、DC-SIGN mRNA与患者临床病理特征相关性。组间比较采用t检验;计数数据以百分率(%)表示,多组间均数比较采用方差分析。结果:肝癌组织样本DC-SIGN蛋白的吸光度值(10.36±1.41)×10^(-3)/μm^(2)、DC-SIGN mRNA表达水平(1.65±0.31)高于癌旁组织样本吸光度值(0.63±0.15)×10^(-3)/μm^(2)、DC-SIGN mRNA表达水平(1.00±0.02,t=48.522、14.796,P<0.05),差异有统计学意义;肝癌组织样本中DC-SIGN蛋白表达与肿瘤TNM分期、包膜存在、血清甲胎蛋白(AFP)指标水平明显相关(t=37.148、17.029、13.894,P值均<0.05);与患者性别、年龄、静脉侵犯、肿瘤分化、肿瘤直径无明显相关(t=0.672、0.603、0.714、0.279、0.530,P值均>0.05);肝癌组织样本中DC-SIGN mRNA表达与肿瘤TNM分期、包膜存在、AFP指标水平明显相关(t=5.726、7.483、2.643,P值均<0.05);与患者性别、年龄、静脉侵犯、肿瘤分化、肿瘤直径无明显相关(t=0.522、0.352、0.443、1.772、1.187,P值均>0.05)。结论:DC-SIGN参与原发性肝癌疾病发生与发展过程。
Objective To analyze the expression and clinical value of dendritic cell-specific intercellular adhesion molecule-3 binding non-integrin(DC-SIGN)in primary liver cancer tissues.Methods From March 2018 to June 2019 in Guizhou Provincial People′s Hospital,40 patients who underwent surgical resection of primary liver cancer were randomly selected.A total of 40 liver cancer tissue samples and 40 cases of adjacent cancerous tissues were collected.The expression of DC-SIGN protein and mRNA in the samples was detected by immunohistochemistry and reverse transcription-polymerase chain reaction(RT-PCR)respectively.SPSS statistical software was used to conduct univariate analysis of the correlation between DC-SIGN in liver cancer tissue samples and clinicopathological characteristics of patients.Results The absorbance values of DC-SIGN protein and mRNA[(10.36±1.41)×10-3/μm2 and(1.65±0.31)]in liver cancer tissue samples were significantly higher than those in the adjacent paracancerous tissue samples[(0.63±0.15)×10-3/μm2 and(1.00±0.02)](t=48.522/14.796,P<0.05).The expression of DC-SIGN protein and mRNA in HCC tissue samples was correlated with pathological characteristics:the expression of DC-SIGN protein in HCC tissue samples was significantly correlated with TNM stage,the presence of tumor capsule and the level of serum alpha-fetoprotein(AFP)index(t=37.148,17.029,13.894,all P<0.05).There was no significant correlation with gender,age,venous invasion,tumor differentiation and tumor diameter(t=0.672,0.603,0.714,0.279,0.530,P>0.05).The expression of DC-SIGN mRNA in HCC tissue samples was significantly correlated with TNM stage,the presence of tumor capsule and AFP index level(t=5.726,7.483,2.643,all P<0.05).There was no significant correlation with gender,age,venous invasion,tumor differentiation and tumor diameter(t=0.522,0.352,0.443,1.772,1.187,P>0.05).Conclusion DC-SIGN is involved in the occurrence and development of primary liver cancer.DC-SIGN can play an important role in the clinical diagnosis and treatment
