百合固金汤治疗非小细胞肺癌的作用机制研究 被引量:5

摘要 目的:非小细胞肺癌是肺癌的一种常见类型,而百合固金汤是目前中药治疗肺癌常见的方剂之一。深入了解百合固金汤治疗非小细胞肺癌的分子机制具有重要的临床意义。方法:我们通过CCK-8和EdU检测百合固金汤对非小细胞肺癌的杀伤作用。接下来,我们在裸鼠背部建立移植瘤模型,从而观察百合固金汤在体内的抑癌作用。进一步,我们通过流式细胞术和蛋白质印迹法初步探索百合固金汤杀伤非小细胞肺癌的生物学机制。结果:我们发现百合固金汤显著减缓肿瘤的生长速度、降低体外克隆形成能力和抑制体内肿瘤生长,且诱导肿瘤细胞在G0G1期发生明显的周期阻滞和凋亡。结论:我们的研究发现百合固金汤能明显抑制非小细胞肺癌细胞的增殖作用并且诱导细胞发生凋亡,从而对非小细胞肺癌产生治疗作用,为促进百合固金汤临床治疗非小细胞肺癌提供了机制证据。 Objective Non small cell lung cancer(NSCLC)is the most common lung cancer,and Baihe Gujin decoction is one of the common prescriptions in the treatment of lung cancer.It has important clinical significance to deeply understand the molecular mechanism of Baihe Gujin Decoction in the treatment of non-small cell lung cancer.Methods We detected the killing effect of Baihe Gujin Decoction on NSCLC by CCK-8 and edu.Next,we established orthotopic transplanted tumor model in nude mice to detect the anti-tumor effect of Baihe Gujin Decoction in vivo.Furthermore,we preliminarily explored the biological mechanism of Baihe Gujin Decoction on non-small cell lung cancer by flow cytometry and Western blot.Results We found that Baihe Gujin Decoction significantly inhibited the growth rate of tumor,decreased the ability of colony formation in vitro and tumor growth in vivo,and induced significant cell cycle G0G1 arrest and apoptosis of tumor cells.Conclusion Our data confirm that Baihe Gujin decoction can significantly inhibit the proliferation of NSCLC cells and induce apoptosis,thus has therapeutic effect on NSCLC,which provides mechanism evidence for promoting clinical treatment of NSCLC with Baihe Gujin decoction.
